| 摘 要： | 目的：探讨三氧化二砷（ＡＳ２Ｏ３）对肺癌细胞株ＮＣＩ－Ｈ６９的增殖抑制和诱导凋亡作用。方法：ＭＴＴ法观察不同浓度ＡＳ２Ｏ３作用不同时间后的ＮＣＩ－Ｈ６９生长的产殖细胞核抗原（ＰＣＮＡ）单抗ＬＳＡＢ免疫组化染色－流式细胞仪检测ＰＣＮＡ阳性细胞百分比；ＦＴＩＣ－ＴＵＮＥＬ法－流式细胞仪检测细胞凋亡指数。结果：随ＡＳ２Ｏ３浓度增加和作用时间延长，ＮＣＩ－Ｈ６９肺癌细胞的细胞活度渐下降。随ＡＳ２Ｏ３浓度增

| Abstract: | Objective Toobserve the effect of proliferatio n inhibition and apop tosis indution of ArsenicTrioxide (AS2O3) on lung neoplasms cell line NCI-H69. Methods MTT assaywas used to observe the proliferation status. Flow cytometry was used to measure thepercentage of proliferation cell nuclear antigen (PCNA) positive cell and apoptosis index.Results AS2O3 could signific antly inhibit th e growth of NCI-H69 cell,this effect was dose and time dependent. The percentag e of PCNA positive cell wasdecreased gradually when concentration of AS2O3 was increased. The apoptosis indexincreased by the increasing of concentration of A S2O3. ConclusionsArsenic Trioxide could inhibit the proliferat ion activity and induce apoptosis of lungneoplasms cell line NCI-H69. It may have potential val ue in the treatment of lung cancer. |
