三七总皂苷对脑出血大鼠细胞凋亡及Bcl-2和Bax表达的影响

摘    要:目的探讨三七总皂苷对脑出血大鼠细胞凋亡及Bcl-2和Bax表达影响。方法将大鼠随机分为假手术组、脑出血组、三七总皂苷治疗组,用大鼠自体血脑内注入法建立脑出血动物模型。造模后72 h采用免疫组化法观察大鼠额顶部皮质Bcl-2和Bax蛋白表达,TUNEL法检测神经细胞凋亡。结果与假手术组比较,脑出血组凋亡细胞明显增加,Bcl-2和Bax表达增加;与脑出血组比较,三七总皂苷治疗组均显著凋亡细胞明显减少,增加Bcl-2表达、减少Bax表达。结论三七总皂苷对脑出血大鼠具有保护作用,其机制可能与通过增加Bcl-2表达,减少Bax表达,及抗凋亡有关。

Effect of Total Panax Notoginseng Saponins on Neural Cell Apoptosis and Expression of Bcl-2 and Bax in Intracerebral Hemorrhage in rats

Abstract:Objective: To study effect of Total Panax Notoginseng Saponins(tPNS) on neural cell apoptosis and expression of Bcl-2 and Bax in intracerebral hemorrhage(ICH) in rats.Methods: In this experiment,rats were randomly divided into 3 groups,which were sham operated group,ICH group,tPNS treated group.Themodel of ICH was established in rats by intracerebral injection of autogenous blood.After 72h of ICH.Neural cell apoptosis was investigated with TUNEL.Expression of Bcl-2 and Bax were also investigated with immunohistochemistry.Result: Compared with sham operated group,neural cell apoptosis increased,expression of Bcl-2 and Bax increased in ICH group.Compared with ICH group,tPNS treated group increased expression of Bcl-2 and reduced expression of Bax.Neural cell apoptosis also reduced.Conclusion: tPNS may reduce intracerebral hemorrhage injure in rats by increasing the expression of Bcl-2 and decreasing the expression of Bax.tPNS plays protective effect from inhibiting cell apoptosis.
