甘草、红景天、黄芪粗提物对中波紫外线诱导小鼠皮肤组织白介素—10的影响

摘    要:目的探索甘草、红景天、黄芪等粗提物对中波紫外线(uw3)损伤BALB/c小鼠皮肤组织的保护作用及其机制。方法将54只BALB/c小鼠用随机数字法分为9组(每组6只):正常对照组、UVB对照组、溶剂对照组、UVB+5%与10%甘草组、UVB+5%与10%红景天组、UVB+5%与10%黄芪组。其中,正常对照组不予处理,UVB对照组单独给予UVB照射,溶剂对照组给予外涂蒸馏水+UVB照射,其余各处理组分别给予外涂相应浓度药物+UVB照射,连续1个月。采用酶联免疫吸附试验(ELISA)测定各组小鼠背部照射部位皮肤组织匀浆上清液中白介素(IL).10水平。结果经UVB慢性照射后小鼠皮肤中Ⅲ—10水平为(838.8±114.34)pg/ml,较正常对照组(568.45±78.8)pg/ml显著增高(P〈0.01),经不同剂量甘草、红景天、黄芪粗提物水溶液处理后可进一步上调IL-10水平,以甘草组最为明显,但IL-10水平与粗提物浓度之间无明显依赖关系。结论在受到UVB慢性照射后小鼠皮肤组织中IL-10表达水平增加,甘草、红景天、黄芪粗提物水溶液可进一步上调其IL-10水平,上述3种药物对UVB诱导的小鼠皮肤损伤的保护作用可能与上调IL-10表达水平有关。

Effect of extractive of Glycyrrhiza, Rhodiola rosea L and Astragalus membrailaceus on IL-10 levels in ultraviolet B irradiated BALB/c mice cutis

Abstract:Objective To investigate the protective effects of topical application of extractive of Glycyrrhiza, Rhodiola rosea L and Astragalus rnernbrailaceus(AM) for ultraviolet B (UVB) irradiated BALB/c mice cutis. Methods 54 BALB/c mice were mndomizely devided into 9 groups (6 mice per group) by stochastic number. Normal contral group, UVB contral group, solution contral group, 5% and 10% Glycyrrhiza plus UVB groups, 5% and 10% Rhodiola rosea L plus UVB groups, and 5% and 10% AM plus UVB groups. Mice in normal contral group didn't accept any treatment, mice in UVB contral group only accepted UVB irradiation, solution control group were given distilled water topically and UVB irradiation. The treatment groups were given homologous medicine topically and UVB irradiation. The mouse in every treatment group were given Glycyrrhiza, Rhodiola rosea L orAM extractive water solution topically on hairless skin of back daily and then irradiated by UVB, once a day for 30 d. Skin samples were taken after 1 m. IL-10 levels in skin samples were measured by ELISA. Results Compared with the control group (568.45〈78.8) pg/ml, IL-10 levels in BALB/c mice skin samples were statistically increased after chronic UVB irradiation (838.8〈114.34) pg/ml (P〈0.01), IL-10 levels were (1319.12〈100.32) pg/ml and (1256.22i169.62) pg/ml in UVB+5% and 10% Glycyrrhiza groups, (1046.07--66.92) pg/ml and (1083.62--117.02) pg/ml in UVB+5% and 10% Rhodiola rosea L groups, (1066.45〈109.58) pg/ml and (1052.9a:54.56) pg/ml in UVB+5% and 10% AM groups respectively, which were evidently higher than UVB group, especially in Glycyrrhiza groups (P〈0.001 vs UVB group, P〈0.05 vs Rhodiola rosea L and AM groups and in Glycyrrhiza groups, P〈0.01 in Rhodiola rosea L and AM groups). However, there were no positive dose-dependent correlationship. Conclusions IL-10 levels in BALB/c mouse skin are increased after UVB irradiation. Topical application of Glycyrrhiza, Rhodiola rosea L and AM can rise regulate IL- 10 levels.
