摘    要:目的:观察体外雪旺细胞环境对大鼠骨髓间充质干细胞(MSCs)的诱导分化作用。方法:分离和体外培养新生大鼠MSCs和雪旺细胞;实验组MSCs经DAPI标记后与雪旺细胞按1∶2比例共培养,观察细胞的形态变化,在第1,3,6天时使用免疫细胞化学染色检测MSCs中Nestin,GFAP及S-100表达。对照组MSCs单独培养,其余处理与实验组相同。结果:共培养1 d后部分MSCs出现雪旺细胞样形态,Nestin表达率为(83.4±3.5)%,随时间增加而降低。GFAP及S-100表达率随时间而增加。对照组细胞形态无变化,不同时间点均有少量MSCs表达Nestin,而GFAP和S-100未见表达。结论:MSCs可通过体外共培养向雪旺细胞分化。

Abstract:Objective: To investigate the inducing effect of Schwann cells environment on the differentiation of bone marrow mesenchymal stem cells(MSCs) in vitro.Methods: MSCs and Schwann cells were isolated from newlyborn rats and cultured in vitro.In treatment group,MSCs were labeled by DAPI and co-cultured with Schwann cells(1∶2 ratio).In control group,MSCs were solely cultured.The morphological change of the cells was observed.The expression of Nestin,GFAP and S-100 was detected by immunocytochemical staining at 1,3 and 6 d after co-culture.Results: The MSCs changed morphologically into Schwann-like cells in co-culture group at 1 d,the positive rate of Nestin was(83.4±3.5)%,then decreased.The expressions of GFAP and S-100 were increased with time.In control group,no morphological change was observed,Nestin expressed at very low level while GFAP and S-100 almost were not expressed.Conclusion: MSCs can be induced to differentiate into Schwann cells by co-culture in vitro.
