Caspase-8在喉癌组织中的表达及其与细胞凋亡相关性研究

目的探讨半胱氨酸蛋白酶-8(Caspase-8)在喉癌组织中的表达及其与细胞凋亡的相关性。方法采用流式细胞术检测45例喉癌患者喉癌组织和相应癌旁组织中Caspase-8的表达,并以20例非喉癌患者喉部正常黏膜组织作对照;同时检测喉癌组织中的细胞凋亡率。结果 (1)喉癌组织中Caspase-8蛋白的表达量及阳性表达率均明显低于癌旁组织及正常喉黏膜组织(P<0.01),癌旁组织及正常喉黏膜组织的表达差异无统计学意义(P>0.05)。(2)Caspase-8蛋白的表达与喉癌组织中的淋巴结转移、临床分期、分化程度有相关性(P<0.05);与临床分型、肿瘤大小、吸烟量、年龄和性别无相关性(P>0.05)。(3)喉癌组织细胞凋亡率与临床分期、淋巴结转移有相关性;与临床分型、病理分级、肿瘤大小、吸烟量、年龄和性别无关。(4)喉癌组织中Caspase-8蛋白表达水平与喉癌组织细胞凋亡率呈正相关(P<0.01)。结论 Caspase-8蛋白的低表达可促进喉癌的发生、发展,并通过抑制凋亡并促进细胞增殖参与喉癌发展。

The Expression of Caspase-8 protein in laryngeal carcinoma and the relation between Caspase-8 and apoptosis

Objective To investigate the expression of Caspase-8 in laryngeal carcinoma and analyze the relation of Caspase-8 and apoptosis.Methods We measured the expression of Caspase-8 protein in 45 laryngeal carcinoma fresh samples and 45 para-carcinoma tissues（to cutting margin0.5cm） and 20 normal laryngeal mucosa samples by flow cytometere（Epics-XLⅡ）,the apoptosis rate was also examined simultaneously.Results（1） The quantitative and qualitative expression of Caspase-8 protein in laryngeal carcinoma tissues is obviously lower than those in para-carcinoma and in normal laryngeal mucosa tissues respectively（P0.01）.There is no significant difference between the expression of para-carcinoma and normal laryngeal mucosa tissues（P0.05）.（2） In laryngeal carcinoma,the expression of Caspase-8 protein wasnt significantly related to patients＇ clinical classification,tumor size,smoking history,patients＇ age and sex（P0.05） but related to metastasis,pathological grade and clinical stage（P0.05）.（3） The apoptosis rate in laryngeal carcinoma wasnt related to patients＇ clinical classification, tumor size,smoking history,patients＇ age and sex and pathological grade,but related to lymph nodes metastasis and clinical stage.（4） In laryngeal carcinoma,there was a positive correlation between the apoptosis rate and FI of Caspase-8（P0.01）.Conclusion The low expression and inhibition of apoptosis of Caspase-8 may contribute to the carcinogenesis and development of laryngeal carcinoma.
