葛脾煎剂调控单核细胞趋化蛋白-1水平逆转早期糖尿病肾病的实验研究

摘    要:目的: 探讨葛脾煎剂对单核细胞趋化蛋白-1(MCP-1)的影响及其对逆转早期糖尿病肾病(DN)的作用。 方法: 选择Wistar雄性大鼠,采用高脂、高糖饮食联合小剂量链脲佐菌素(STZ)腹腔注射诱导的STZ糖尿病大鼠共41只,随机分为STZ糖尿病大鼠对照组14只,中药(葛脾煎剂)治疗组13只,西药(苯那普利)治疗组14只,分别观察8周;检测各组体重、肾重指数、血糖、尿微量白蛋白、血尿MCP-1、血8-异前列腺素 F2α(8-iso-PGF2α)、肿瘤坏死因子-α(TNF-α),并进行肾脏病理学检查。 结果: 中药治疗组肾脏重量和肾重指数低于对照组( P <0.05),UAER、血尿MCP-1、血8-iso-PGF2α和TNF-α均明显低于对照组( P <0.01);而两治疗组,对上述指标的影响基本类似,两组间无明显差异( P >0.05)。另外,各组8-iso-PGF2α、MCP-1、TNF-α均与UAER呈正相关关系。中药葛脾煎剂能减轻肾脏肥大及肾小球系膜基质增生、硬化。 结论: 糖尿病肾病有自身免疫性炎症和氧化应激反应参入,中药葛脾煎剂对多种炎症因子具有抑制作用,从而可逆转早期糖尿病肾病。
