荷叶碱降尿酸的1H-NMR代谢组学研究

摘    要:目的] 研究荷叶碱对高尿酸血症小鼠的作用,筛选与高尿酸血症代谢有关的生物标志物。方法] 采用小鼠长期给药氧嗪酸钾模型,连续给药14 d后,运用核磁共振氢谱(1H-NMR)技术同时检测高尿酸血症小鼠的血浆和尿液样本。结果] 通过1H-NMR分析方法在血浆和尿液样品中共筛选出9个潜在生物标志物。结论] 荷叶碱对高尿酸血症小鼠的代谢组学研究表明高尿酸血症的病理过程主要涉及到酪氨酸代谢、柠檬酸代谢、精氨酸与脯氨酸代谢以及糖酵解和糖异生代谢途径,预示着这些潜在生物标志物不仅在早期疾病诊断中起重要作用,而且在预防高尿酸血症中也有重大意义。

Abstract:Objective] To study the effect of nuciferine on hyperuricemia mice, and to screen for biomarkers related to hyperuricemia metabolism.Methods] A long-term administration model for mice was used. After continuous administration for 14 days, proton nuclear magnetic resonance(1H-NMR) was used to detect plasma and urine samples from hyperuricemia mice.Results] A total of 9 potential biomarkers were screened from plasma and urine samples by 1H-NMR analysis.Conclusion] The metabolomics study of nuciferine in hyperuricemia mice further illustrates that the pathological process of hyperuricemia mainly involves tyrosine metabolism, citric acid metabolism, arginine and proline metabolism, and glycolysis and the gluconeogenesis pathway, which indicates that these potential biomarkers not only play an important role in early disease diagnosis, but also have great significance in the prevention of hyperuricemia.
