中药对热适应大鼠肝与心肌细胞HSP70表达影响的研究

探讨中药 (主要成分为红参、黄芪、红景天等 )对热适应不同阶段HSP70表达的影响 ,为用药物提高热区部队热适应能力提供实验依据。采用人工热气候室 ( 3 4± 1℃、RH60 % ) ,建立热适应动物模型。SD大鼠 160只分为对照组、热适应组、中药对照组、中药热适应组 ,每组又分为热适应 2、7、14、2 8天 4个时段。采用免疫组化SP法染色 ,应用图像分析系统分析肝、心肌组织HSP70含量的变化。结果 ,在热适应的 4个阶段 ,肝与心肌细胞的HSP70表达强度变化呈现如下趋势 :中药热适应组 >热适应组 >对照组和中药对照组 (P <0 0 1或 0 0 5 ) ;热适应不同时期中药热适应组肝与心肌HSP70表达强度无明显变化 ;适应 2天时HSP70在核内表达强于胞浆 ,而此时中药组则是胞浆表达强于胞核 ;热适应 2天与 2 8天时血清中肝与心肌细胞内酶浓度明显增高 ,而中药热适应组血清酶浓度明显低于热适应组。中药可显著增强热适应不同阶段肝与心肌细胞HSP70的表达强度 ,减轻热适应时肝与心肌细胞的损伤 ,使机体较早达到热适应状态。同时中药的细胞保护作用机理也可能与增强HSP70的高表达有关。

Effects of the Chinese traditional medicine on expression of HSP70 in the cells of heat acclimation rat liver and cardiac muscle

Objective :To observe the changes of HSP70 expression in different heat acclimation stages,and to get the evidence of the increased heat adaptation of organism in hot environment with the usages of the Chinese traditional medicine. Method :The heat acclimation model was established in rats with the artificial hot climatic chamber(34±1℃?rh 60%).160 healthy SD rats were distributed randomly into control group(C),heat acclimation group(A),Chinese traditional medicine control group(DC),and Chinese traditional medicine heat acclimation group(DA),and into subgroups including 2d,7d,14d,28d stages from each one of the groups.The immunocyto chemistry was used to detect HSP70 expression in rat liver and cardiac muscle,and photography analytic software was used to analyze HSP70 expression. Result :1)The expression intensity of HSP70 in various groups is as follows:DA>A>C or DC( P <0 01 or 0 05)during all stges of the heat acclimation.2)There is no significant difference in expression intensity of HSP70 among the various stages of the heat acclimation.3)After the heat acclimation 2d,the HSP70 expression in cell nuclei of the liver and cardiac muscle cells is stronger than that in cytoplasm in A group,and inverse in DA group.4)The activity of ALT,AST,CK and HBD showsan increase in heat acclimation 2d and 28d in A groups,but not so high in DA groups. Conclusion :Chinese medicine can increase the HSP70 expression,which results in a decreasement of membrane damaged and a rapid tolerance to heat.It suggests that a protective effect on heat is relative to HSP70.
