急性心肌缺血损伤后大鼠经穴CaM含量的变化及不同频率电针的干预作用

摘    要:目的观察大鼠急性心肌缺血损伤(acute myocardial ischemia,AMI)后,经穴钙调蛋白(Calmodulin,CaM)含量的特异性变化及不同频率电针"内关"穴在保护急性心肌缺血损伤时对相关经穴CaM的特异性影响。方法 30只雄性Wistar大鼠随机分为4组,正常组6只,将18只心肌缺血损伤模型成功大鼠平均分为模型组6只、低频治疗组6只和高频治疗组6只,采用酶联免疫吸附试验检测CaM含量。结果心肌缺血损伤后,大鼠相关经脉的"内关"穴区和"天泉"穴区、或非相关经脉"足三里"穴区的CaM水平较正常组显著升高(P<0.05),而低频电针治疗后3个穴区CaM含量显著下降(P<0.05),高频电针治疗后"天泉"或"足三里"穴区CaM含量显著下降(P<0.001或P<0.05)。结论低频或高频电针"内关"穴可保护大鼠心肌缺血损伤,其机制可能与减少心脏相关的经脉穴位"内关"和"天泉"穴区、非相关经脉"足三里"穴区CaM含量有关;低频或高频电针对心肌缺血损伤大鼠相关穴区CaM影响的机制值得探讨。

Abstract:Objective To observe the specific changes of calmodulin(CaM) content around acupoints and the specific influences of electroacupunture in different frequency on CaM content around acupoint when electroacupunture was used in Neiguan(PC6) in rats with acute myocardial ischemia(AMI).Methods Male Wistar rats(n=30) were randomly divided into four groups.There were 6 rats in normal group,and other successfully modeled rats(n=18) were divided into model group,group treated with low-frequency electroacupunture(low-frequency group) and group of high-frequency electroacupunture(high-frequency group,each n=6).The content of CaM was detected by using enzyme-linked immunosorbent assay(ELISA).Results After the injury of AMI,CaM content increased significantly around PC6 and Tianquan(PC2) in relative meridian and around Zusanli(ST36) in irrelative meridian compared with normal group(P<0.05),while CaM content decreased significantly around the above three acupoint areas after the treatment with low-frequency electroacupunture(P<0.05).The content of CaM decreased significantly around PC2 and ST36 after the treatment with high-frequency electroacupunture(P<0.001 of P<0.05).Conclusion Low-frequency electroacupunture or high-frequency electroacupunture used in PC6 can protect rats from the injury of AMI.The mechanism may be related to the decrease of CaM content around PC6 and PC2 in heart-related meridian and around ST36 in irrelative meridian,which is worth further study.
