组蛋白去乙酰化酶抑制剂曲古抑菌素对原发性肝癌细胞周期和凋亡的影响

目的 观察组蛋白去乙酰化酶抑制剂曲古抑菌素(TSA)对原发性肝癌细胞周期和凋亡的影响.方法 采用Wester blot检测经TSA处理后肝癌细胞的乙酰化水平;运用流式细胞计数(FCM)分析TSA对肝癌细胞周期和凋亡的影响.结果 TSA处理BEL-7402细胞16 h后细胞乙酰化水平到达最高峰;用TSA处理BEL-7402细胞16 h后,肝癌细胞G0/G1细胞从43.39%升高到53.82%,统计学检验,其差异有统计学意义.S细胞从26.62%降到20.85%,G2/M细胞从27.86%降到22.07%,细胞凋亡由0.93%降到0.75%,差异无统计学意义(P>0.05).结论 TSA能够阻滞HCC细胞周期C1～S期进程,抑制细胞生长,而对HCC细胞凋亡无明显影响.

Effects of histone deacetylase inhibitors TSA on hepatocellular carcinoma cells cycle and apoptosis

Objective To study the effects of histone deacetylase inhibitors TSA on hepatocellular carcinoma (HCC) cells cycle and apoptosis. Methods The changes in cell cycle, apoptosis and cyclin were detected by flow cytometry. Results After treatment with TSA,the ratio of G0/G1 was increased from43.39% to 53.82% ( P < 0.05 ), while percentage of S cells, G2/M cells, and apoptosis were decreased from 26.62% to 20.85%, from 27.86% to 22.07% and from 0. 93% to 0.75%, respectively (P >0.05). Conclusion After treatment with TSA, the G1-S cell cycle progression of HCC ceils can be blocked, the growth of HCC was suppressed, and the apoptosis of HCC cells had no significant changes.
