牛蒡苷元对自发性高血压大鼠的降压及对靶器官的保护作用

摘    要:目的研究牛蒡苷元(ARC—G)对自发性高血压大鼠(SIqR)和正常Wistar大鼠血压的调节、靶器官的保护作用。方法12周龄的SHRs(280g~320g)40只随机分为牛蒡苷元高、中、低剂量组(100mg/kg、50mg/kg、25mg/kg),硝苯地平(3mg/kg)阳性对照组,溶剂对照组;相应周龄的Wistar大鼠16只,随机分为牛蒡苷元(100mg/kg)和溶剂对照组。所有溶剂均为1%羧甲基纤维素,灌胃给药8周。观察牛蒡苷元对SHR和Wistar大鼠血压、心、肾高血压主要靶器官质量指数的影响。结果牛蒡苷元对各组大鼠体重无影响,但可剂量依赖性地降低SHR大鼠血压;牛蒡苷元和硝苯地平能明显抑制SHR左心室肥厚,SHR各组的SBP和左室质量指数、肾脏质量指数均明显高于同龄Wistar大鼠耿0.01)。给药治疗2周后,与SHR对照组比较ARC—G高中剂量组可明显降低SBP(P〈0.05或尺0.01),可明显降低左室质量指数(P〈0.05或P〈0.01);对肾脏质量指数无明显影响(P〉0.05);牛蒡苷元对Wistar大鼠各指标均无明显影响,与对照组相比(P〉0.05)。结论牛蒡苷元对SHR有降压及逆转左心室肥厚的作用。

Abstract:Objective To investigate hypotensive and target organs protective effecs of long-term treatment with aretigenin (ARC-G). Methods Forty 12-week-old male SHR(280-320g) were randomly assigned to five groups (aretigenin 100mg/ kg, arctigenin 50mg/kg, arctigenin 25mg/kg nifedipine3mg/kg, SHR vehicle) and matched Wistar rats were randomly assigned to two groups (Wistar+aretigeninlO0mg/kg and Wistar vehicle). All animals were treated for 8 weeks. Systolic blood pressure was determined every other week. Bodyweight was determined every week. Cardiac and renal morphology were examined. Results Chronic arctigenin treatment reduced systolic blood pressure in a dose dependent manner, and significantly reduced left ventricular hypertrophy. The efficianey of arctigenin (50,100mg/kg) is similar to nifedipine. Conclusion Aretigenin can decrease high blood pressure and reverse left ventrieular hypotrophy in SHR.
