硒与甲基汞对体外培养细胞SCE的作用

摘    要:目的探讨甲基汞对染色体的毒性,解释其遗传毒性,探讨硒对甲基汞引起的细胞毒性的保护机制。方法采用正常人外周血淋巴细胞体外培养。分为三个大组,每组给于不同剂量水平和施加不同的因素:第一组为甲基汞作用组,分为6个亚组5个剂量水平,加入甲基汞的终浓度为1×10-9~1×10-6mol·L-1;第二组在加入甲基汞的基础上加入亚硒酸钠,同第一组一样分为6个亚组5个剂量水平,其亚硒酸钠浓度为1×10-7~1×10-5mol·L-1;第三组只加入亚硒酸钠,其浓度为1×10-7~1×10-5mol·L-1。37℃±0.5℃培养72h,制片染色,采用双盲法,观察20~30个中期细胞的姊妹染色单体交换(SCE)。结果亚硒酸钠能使由甲基汞引起的细胞遗传损伤明显减轻,经统计处理P<0.01。结论甲基汞可引起体外培养的正常人的外周血淋巴细胞SCE的增加。亚硒酸钠能在一定程度上减轻由甲基汞引起的细胞遗传物资的损伤。

Abstract:Objective To research the toxicity of methylmercury for chromosome and heredity, and investigate the protective effects of selenium on cytotoxicity. Method The lymphocyte of the peripheral blood of normal people were cultured outside body and were divided into three groups .Each group was given different dose level and different factor. Group 1 was the group of Methylmercury action and was divided into 6 sub-groups with 5 different dose levels. The concentration of methylmercury is 1×10 -9 to 1×10 -6 mol·L -1 . In groups 2 selenium selenite was put in addition to methylmercury and it was also divided into 6 sub- groups with 5 different dose levels. The concentration of sodium selenite is 1×10 -7 to 5×10 -5 mol· L -1 . In groups 3 only sodium selenaite was put ,its concentration was 1×10 -7 to 1×10 -5 mol·L -1 .All the above were Culture for 72 h.37℃±0.5℃. and were made into sections and SCE of 20~30 mid-phase cells were observed with double blind method. Result Sodium selenite can make cytogenetic injured due to methylmercury reduced greatly. P <0.01.after significant test. Conclusion Methylmercury can lead to increase of SCE of cells of peripheral blood lymphocyte of normal people cultured outside body. Sodium selenite can reduce to some degree the injury of cytogenetics matters due to methylmercury.
