砷剂诱导细胞凋亡机制及与维生素C联合作用的研究进展

摘    要:砷剂在治疗血液系统恶性肿瘤方面有较好的疗效,它诱导恶性细胞凋亡的机制主要包括砷剂可以使肿瘤细胞的PML-RARa蛋白降解,影响细胞内的信号传导通路,改变线粒体的膜通透性。新的研究资料表明砷剂可以影响细胞线粒体的电子传递链,改变线粒体膜电位,影响细胞内的ROS水平而诱导细胞凋亡。维生素C是一种传统的还原剂有时对细胞又起着氧化剂的作用。本文就此做一些总结,并对砷剂和维生素C的联合应用的机制进行一些探讨。

Abstract:As a drug, arsenical possessing a long history with recent combination use of vitamin C for treating hematologic malignancies, has achieved good efficacy clinically. But the apoptotic mechanism of the collaboration is still in study. The past studies about arsenicals revealed the capability of inducing the degradation of PML-RARa protein in tumor cells, influencing the signal transmitting pathway, and changing the permeability of mitochondria membrane. The recent studies show that arsenicals can interfere the electron transmit chain on mitochondria, change the membrane voltage of mitochondria, influence the level of ROS in the cell, and so induce cell apoptosis. Vitamin C is a conventional de-oxidant, but some experiments show it to be an oxidant to cell. So this article will discuss the possible mechanism of the combination use of the two drugs.
