海风藤酮对小鼠抗着床作用的细胞化学和电镜观察

摘    要:目的观察海风藤酮对小鼠的抗着床作用及其细胞化学和超微结构的变化。②方法向妊娠第2天小鼠子宫角内注射不同剂量的海风藤酮,于妊娠第8天和第10天观察其对滋养层细胞的刀豆凝集素(ConA)受体和麦胚凝集素(WGA)受体表达,以及对滋养层细胞和子宫蜕膜细胞超微结构的影响。③结果随着海风藤酮剂量增大,滋养层细胞的WGA和ConA受体的阳性反应减弱,以WGA受体的改变更明显。电镜所见,实验组绒毛浅层滋养层细胞胞浆电子密度低,核固缩;深层滋养层细胞溶酶体增多,见髓样小体,子宫蜕膜的大蜕膜细胞胞浆溶酶体增多,胞浆空泡化,核膜溶解。④结论海风藤酮可使小鼠胚胎滋养细胞的WGA和ConA受体糖蛋白减少,从而干扰了细胞之间的识别黏附,破坏了胚胎正常生长和发育的内环境,抑制着床过程。

Abstract:Objective To investigate the effect of Kadsurenone on the implantation of mouse embryos and to observe the cytochemical and ultra structural changes. Methods On the day 2 of pregnancy, Kadsurenone in different dosages was intrauterohornly injected into the mice. On the day 8 and day 10 of pregnancy, the expression of WGA and ConA receptors as well as ultrastructural changes in trophoblastic cells and decidual cells were observed. Results Cytochemical observations showed that the positive reaction of WGA and ConA receptors of trophoblastic cells weakened along with the increasing dosages of Kadsurenone, notablly with the WGA staining. Ultrastructurally, the cytoplasms of trophoplastic cells within the superficial area in the experimental groups became less dense, nuclei pyknosed; while in the deeper area, the lysosomes in the trophoblastic cells remarkablly increased in number, Even the myelinated bodies were found in these cells. In larger decidual cells, the lysosomes were also greatly increased accompanied by vacuolation of the cytoplasm and rupture of the nuclear membrane. Conclusions By means of decreasing the amount of glycoprotein in WGA and ConA receptors in trophoblastic cells, Kadsurenone can interfere with the intercellular adherence and attachment, destroy the enviroment where the embryos survived and developed and inhibit the process of implantation.
