TNF-α含量的改变与肾小管细胞缺氧-复氧性损伤关系的研究

摘    要:目的:探讨TNF-α含量的改变与肾小管上皮细胞株(HK-2)缺氧-复氧性损伤的关系。方法:以HK-2细胞株作为研究材料,采用液体石蜡覆盖法建立缺氧损伤模型,分别在缺氧4、12、24h和复氧4、12、24h,采用放射免疫分析法(RIA)、生化法和台盼蓝拒染法,检测培养液中TNF-α的含量、乳酸脱氢酶(LDH)的活性、细胞计数及活细胞率的改变。结果:缺氧后4h,TNF-α的含量、LDH的活性开始升高,细胞总数及活细胞计数开始下降,并于缺氧后24h达高峰。TNF-α含量的改变与LDH活性的升高呈显著的正相关(r=0.89,P<0.05),与活细胞计数的下降呈显著的负相关(r=-0.91,P<0.05)。结论:肾小管缺氧-复氧后,TNF-α的产生增多,并参与了肾小管细胞损伤的过程。

Abstract:AIM: To investigate the relationship between TNF-alpha and renal tubular cell injury caused by anoxia/reoxygenation. METHODS: Human renal proximal tubular cell line HK-2 was used as model. Anoxia/reoxygenation were produced by covering/de-covering the cell culture with liquid paraffin wax. The level of TNF-alpha and the activity of lactate dehydrogenase(LDH) in the culture medium was determined by radioimmunoassay(RIA) and biochemical methods, respectively. Trypan blue exclusion was used to measure cell viability. RESULTS: Anoxia/reoxygenation could increase TNF-alpha level and LDH activity, but decrease viability of HK-2 cells. TNF-alpha level was positively correlated with LDH activity (r=0.89, P<0.05) and negatively with the cell viability (r=-0.91, P<0.05). CONCLUSION: TNF-alpha induced by anoxia/reoxygenation may participate in the process of renal tubular cell injury.
