iNOS mRNA在缺血再灌注损伤鼠视网膜中的表达

目的　研究iNOSmNRA在缺血再灌注过程鼠视网膜中的表达 ,探讨NO对视网膜缺血再灌注损伤的作用和意义。方法　动物模型采用升高眼压造成视网膜缺血 ,再恢复正常眼压形成血流再灌注。用Biotin标记iNOScRNA探针进行分子原位杂交。结果　正常组、对照组视网膜没有iNOSmRNA表达 ;再灌注 3、12、2 4h均有iNOSmRNA表达 ,与正常组相比差异有显著性 (P <0 0 1) ,并且再灌注 12hiNOSmRNA表达量最高 ,明显高于其他实验组 (P <0 0 1) ;再灌注48、96h没有发现iNOSmRNA表达。结论　在缺血再灌注过程视网膜有较高的iNOSmRNA表达 ,iNOS催化产生的NO可能参与了视网膜的缺血再灌注损伤。

Expression of inducible nitric oxide synthase mRNA in rat retinal ischemia-reperfusion injury

ObjectiveTo investigate the role and siginificance of NO in the retinal ischemic disease by studying gene expression of iNOS after the rats retinal ischemia-reperfusion injury.MethodsModel were created by raising the intraocular pressure to form retinal ischemia,then reperfusion was done by lowering the intraocular pressure to normal.The expression of iNOS was determined by in situ hybridization of iNOS cRNA probe labeled with biotin.ResultsThe expression of iNOS mRNA was not found in normal group and control group,but it was determined at 3,12 and 24 hours of reperfusion after 2 hours of ischemia,with peak expression at 12 hours after reperfusion (P<0.01).There were significant difference in comparison with the normal group(P<0.01).But no expression of iNOS mRNA was found at 48 and 96 hours after reperfusion.ConclusionsThe results suggest that nitric oxide produced by iNOS may mediate retinal ischemia-reperfusion injury.
