降糖舒心颗粒对晚期糖化终产物致离体血管平滑肌细胞增殖的影响

摘    要:目的:观察降糖舒心颗粒对晚期糖化终产物(AGEs)致离体血管平滑肌细胞(VSMCs)增殖的影响.方法:分离正常大鼠胸主动脉VSMCs,并传代培养,用AGEs造成胸主动脉VSMCs增殖模型.给正常大鼠灌胃降糖舒心颗粒混悬液10d,制备含药血清,采用噻唑蓝(MTT)比色法和3H-TdR掺入试验测定其吸收度和3H-TdR掺入量(cpm值).结果:AGEs能明显促进VSMCs增殖,降糖舒心颗粒含药血清能抑制胸主动脉VSMCs的增殖及3H-TdR掺入(与对照组比P<0.05或P<0.01),并呈现一定的量效关系.结论:降糖舒心颗粒对AGEs诱导VSMCs的增殖有一定的抑制作用.

Abstract:Objective:To investigate the effect of Jiangtang Shuxin Keli (JSK) on proliferation of vascular smooth muscle cells (VSMCs) induced by advanced glycosylation endproducts (AGEs) in vitro.Methods: VSMCs isolated from thoracic aorta of the healthy rats were passage-cultured. The medicated serum was prepared by feeding the healthy rats with JSK suspension by gastrogavage for 10 days. After being cultured with AGEs and JSK,respectively, the absorbance of VSMCs was measured by methyl thiazolyl tetrazolium colometry,and incorporation of 3H-TdR (cpm) into VSMCs was quantified by a radioactive assay. Results: Compared to the control group, significant increments in the absorbance of VSMCs and the 3H-TdR incorporation were shown in the AGEs group (P < 0. 05 ), and significant declines in the absorbance of VSMCs and the 3H-TdR incorporation in the serum containing JSK metabolites group (P<0.01) in a dose-dependent manner. Conclusion: JSK works by inhibiting nonenzymatic glycosylation that leads to block the proliferation of VSMCs.
