原发性肝细胞癌中乙型肝炎病毒X抗原表达的分析

我们前已报道用抗ＨＢＶｘ抗原（ＨＢｘＡｇ）三种不同肽段的抗体检出肝癌细胞中ＨＢｘＡｇ的存在。我们用ＰＣＲ技术扩增肝癌肝组织中的ｘ基因，用重组ＨＢｘＡｇ及其抗体分别检测肝癌患者血清中的抗ＨＢｘ和ＨＢｘＡｇ并用免疫组织化学的方法分析肝组织中ＨＢｘＡｇ的表达及其分布。我们发现ＨＢｘＡｇ在肝癌肝组织中阳性率高达９２．６％而且分布广泛，主要表达在细胞浆，但细胞核内亦有见到。癌旁组织检出率也很高，而且在小胆管上皮细胞内亦见有ＨＢｘＡｇ的存在。研究结果表明ＨＢｘＡｇ的表达与肝癌有密切关系

ANALYSIS OF HBV X ANTIGEN EXRESSION IN HEPATOCELLULAR CARCINOMA

In our previous papers intrahepatic localization of HB xAg in liver cancer was investigated usingantibodies against peptide fragments representing three particular regions of X gene sequence. In thisstudy PCR technique was used to detect the X gene in the liver tissue sections.The antisera against therecombinant HBxAg which was synthesized in E. coli were prepared andused for the detection ofHBxAg in the liver cell of 7 8 patients with HCC. And the HBxAg/anti HBx were also analizedin the sera from the patients using ELISA. It was found that HBxAg was not only seen in hepatocyte cyto-plasm but also in nuclei and the total positivity rate of HBxAg is 92.6%(75/78). Besides this, we alsofound the predominant expression of HBXAg in bile duct ethelial cells. Our findings show that theexpression of HBxAg closely related to the development of HCC.
