摘    要:目的研究大蒜素对人肝癌HepG2细胞增殖抑制、细胞凋亡及细胞周期的影响。方法用不同浓度的大蒜素作用于体外培养的HepG2细胞,采用MTT法检测大蒜素对HepG2细胞增殖抑制能力,荧光显微镜观察HepG2细胞的形态学变化,流式细胞术检测大蒜素对HepG2细胞周期和凋亡率的影响。结果大蒜素可抑制人肝癌HepG2细胞增殖,且具有明显剂量和时间依赖性,24h、48h和72h大蒜素抑制HepG2细胞的IC50分别为(48.26±1.66)μg/mL、(31.89±1.51)μg/mL和(23.79±1.32)μg/mL。32μg/mL大蒜素作用HepG2细胞48h,可诱导其产生明显的细胞凋亡特征。流式细胞术检测表明大蒜素作用HepG2细胞后,可将细胞阻滞在G2/M期。结论大蒜素对人肝癌HepG2细胞增殖具有抑制作用,可能与诱导细胞凋亡和阻滞细胞周期有关。

Inhibitive Effect of Allicin on Proliferation of Human Hepatoma Cell HepG2

Abstract:Objective To investigate the effect of allicin on cell growth, cell apoptosis and cell cycle in human hepatoma cell HepG2 in vitro. Methods The human hepatoma cell HepG2 was cultured and treated by alliein in different concentrations in vitro. MTT assay was used to test dynamically the inhibitive effect of the cell growth. Morphologic change was observed by using the fluorescent microscope. The modulation of DNA cell cycle and apoptosis induction were measured by flow eytometry. Results Alliein had inhibitive effects on growth of HepG2 cells in a dose and time-dependent manner, with the IC50 value of(48.26 ±1.66)μ g/mL, (31.89 ± 1.51 )μg/mL and(23.79 ±1.32 )μ g/mL at 24h, 48h and 72h respectively. When the cells were treated with allicin at the concentration of 32μ g/mL for 48h, HepG2 cells were notably induced into apoptosis. Flow eytometry showed that allicin could block cell growth at G2/M phase. Conclusion It is suggested that allicin inhibits HepG2 cell proliferation, which is associated with the induction of apoptosis and arrestment of cell cycle.
