硫酸镁对兔视网膜缺血再灌注损伤的保护作用

摘    要:目的探讨硫酸镁对实验性兔视网膜缺血再灌注损伤后的保护作用及其机制。方法用眼内灌注法制成视网膜缺血再灌注模型。32只健康成年新西兰大白兔随机分为对照组、模型组、硫酸镁治疗A组和硫酸镁治疗B组。硫酸镁治疗A组于缺血后立即按140 mg.kg-1耳缘静脉推注2 mol.L-1硫酸镁,随后腹腔内补充注射1 g;硫酸镁治疗B组于缺血后,按600 mg.kg-1腹腔内注射2 mol.L-1硫酸镁。各组分别于再灌注6 h后空气栓塞法处死兔,经光镜、电镜观察视网膜的改变。结果再灌注6 h后,光镜下模型组视网膜神经节细胞层、内核层变薄,电镜下出现核固缩,线粒体空泡化。经硫酸镁治疗后视网膜神经节细胞层、内核层的形态学明显改善,且硫酸镁治疗A组视网膜神经节细胞层和内核层损害较硫酸镁治疗B组明显减轻。结论硫酸镁可能在治疗视网膜缺血性疾病中有一定应用价值。

Abstract:Objective To investigate the protective effect of magnesium sulfate ( MgSO4 ) on retina of rabbit with ischemia-reperfusion injury. Methods The model of experimental retinal ischemia-reperfusion injury was created by intraocular perfusion. Thirty-two rabbits were randomly divided into model group, control group and MgSO4 treatment group A and MgSO4 treatment group B. MgSO4 treatment group A was given 2 mol · L^-1 MgSO4 ( 140 mg · kg^-1 ) through intravenous injection postischemia,then 1 g MgSO4 was given to them by intraperitoneal injection. MgSO4 treatment group B was given 2 mol · L^-1 MgSO4 (600 mg· kg^-1) through intraperitoneal injection postischemia. Repeffusion for 6 hours,then killed the rabbits with air embolism and observed the changes of retina by light microscope and electron microscope. Results Stratum ganglionare nervi optici and inner nuclear layer became thinner under light microscope, and there was pyknosis and mitochondria vacuolization under electron microscope after reperfusion for 6 hours. The morphology of stratum ganglionare nervi optiei and inner nuclear layer improved obviously after treatment with MgSO4, and the injure of MgSO4 treatment group A was lower than MgSO4 treatment group B. Conclusion Magnesium sulfate may have some applied value for treating the disease of retina ischemia.
