人类结肠癌单克隆抗体的研究

用大肠癌HT29作抗原免疫小鼠,取其脾脏的淋巴细胞与骨髓瘤细胞融合,产生三株杂交瘤细胞——Ga_3、Gb_6和Cb-5,其所作用的抗原为肿瘤相关抗原。间接免疫荧光的研究发现:三株抗体与肿瘤的石腊切片无反应,但与冰冻切片及大肠癌细胞株作短期固定者反应阳性。Ga_3属IgG_(2a),Gb_6和Cb_5属IgM。三株抗体可能有助于结肠癌的免疫诊断。

MONOCLONAL ANTIBODIES OF HUMAN COLONIC CANCER

Three monoclonal antibodies,Ga3,Gb6 and Cb5,were produced with spleen cells from mice immunized against the colonic cancer cell line HT29. Using ELISA screening test,Ga3 reacted with 5/8,Cb5 with 6/8,and Gb6 with all of the cell lines tested. Five pancreatic cancer cell lines were also tested by these monoclonal antibodies. Three of them were negative,one was positive to all monoclonal antibodies,one was positive to Ga3,Cb5 and negative to Gb6.Gb6 also reacted wiht stomach cancer with which the others did not. Although Ga3 did not react with colonic cancer tissue using paraffin embedding method,it did react with colonic cancer cell which was freshly prepared and fixed with acetone. Competition studies suggest that Ga3 is different from other monoclonal antibodies in epitope bindings, it belongs to IgG2a class, it may be useful in cancer diagnosis and therapy.
