HPV16 E6、P21^WAF1、CDK2、Livin在宫颈癌中的表达及其意义

摘    要:目的探讨人乳头瘤病毒(HPV)16E6、P21^WAF1、CDK2、livin在宫颈癌中的表达及P21^WAF1、CDK2、livin与E6的关系。方法采用免疫组化SP法检测HPV16E6、P21P21^WAF1、CDK2、livin在20例宫颈癌、40例宫颈上皮内瘤变(CIN)及20例正常对照组中的表达。应用图像分析软件分别对此四个因子的表达进行平均光密度分析。结果E6、P21^WAF1、CDK2、livin蛋白于宫颈癌组高于CIN1-2组及正常组(P〈0.05),CIN3组高于CIN1-2组及正常组(P〈0.05),宫颈癌组与CIN3组之间差异无统计学意义(P〉0.05),CIN1-2组与正常组之间差异无统计学意义(P〉0.05)。E6分别与P21^WAF1、CDK2、livin成正相关(相关系数r分别为0.706、0.713、0.711,P〈0.05)。结论E6、CDK2与宫颈癌的发生、发展密切相关,E6、CDK2异常高表达可能是宫颈疾病恶变早期事件。livin可能成为宫颈癌潜在的治疗分子靶向。

Abstract:Objective To investigate the expression of human papillomavirus (HPV) 16 E6, P21^WAF1, CDK2 and livin in cervical cancer and further to explore the correlation between P21^WAF1, CDK2, livin and E6. Methods Immunohistochemical SP method was used to detect HPV16 E6, P21^WAF1, CDK2 and livin proteins in 20 cases of cervical cancer (CC) , 40 cases of cervical intraepithelial neoplasia (CIN) and 20 cases of normal controls. The mean density (MD) of the expression of E6, P21^WAF1, CDK2 and livin were counted, respectively, with image analysis software. Results The levels of expression of E6, P21^WAF1 , CDK2 and livin were all significantly higher in CC than that of in CIN1-2 and normal controls (P 〈 0.05) , and their levels were also higher in CIN3 than that of in CIN1-2 and normal controls (P 〈0.05). However, the levels were not significantly different between CIN1-2 and normal controls, and also not significantly different between CC and CIN3. The expression of E6 was correlated with P21^WAF1, CDK2 and livin, respectively (P 〈 0.05) , and their correlation coefficients were 0. 706, 0. 713 and 0. 711, respectively. Conclusions E6 and CDK2 play an important role in the occurring and development of CC. Livin may become an ideally molecular target in the treatment of CC.
