摘    要:目的观察小鼠脾区经532 nm激光照射后免疫功能的改变。方法用532 nm连续激光辐照KM小鼠脾区10 min/d,连续4 d,照射功率分别为0 mW(对照组)、20 mW、50 mW。观察照完后1 d腹腔巨噬细胞吞噬指数、胸腺和脾脏指数的改变; 用流式细胞仪检测脾细胞活性氧以及脾脏、外周血中CD3^+、CD4^+和CD8^+T细胞亚群的变化。结果(1)与对照组比较,经20 mW照射后脾脏指数明显升高,而50 mW组虽高于对照组但未见明显差别; 胸腺指数照射后未见明显改变。(2)照射后腹腔巨噬细胞吞噬指数较对照组明显升高,20 mW组(6.46±1.30)和50 mW组(7.13±1.03)分别为对照组(4.97±0.72)的1.30和1.43倍。(3)小鼠脾细胞活性氧水平随照射剂量增加而降低,50mW组活性氧水平明显低于对照组。(4)经20或50 mW照射后脾脏CD3^+、CD8^+T细胞明显降低,外周血CD4^+/CD8^+比值出现降低(P〈0.05)。结论小鼠经低能量的532 nm激光照射脾脏后,其生物调节效应,不仅仅是刺激作用,也可能是抑制效应,具有一定的双重调节性。

Abstract:Objective To observe the changes of mouse immune function after 532 nm laser irradiation Methods Kunming mouse spleen area was exposed to 0, 20 and 50 mW 532 nm laser irradiation for 10 min, once a day for 4 days. The phagocytosis index of one day postirradiation mouse peritoneal macropha- ges, and the thymus and spleen indexes were analyzed, the reactive oxygen species (ROS) in spleen,and T cell subpopulations of CD3^+ ,CD4^+ and CD8^+ in peripheral blood and spleen were measured by flow eytometry. Results ( 1 ) At 1 day after irradiation the spleen index of 20 mW group was significantlyhigher ( P 〈 0.05 ) , while the spleen index in 50 no statistically significant changes were observed, mW group and thymus index in 20 and 50 mW groups when compared with that of the control group. (2) The phagocytosis index of peritoneal macrophages was higher than that of the control group ( P 〈 0.01 ) , phagocytosis index in the 20 mW group (6.46 ± 1.30) and 50 mW group(7.13 ± 1.03 ) were approximately 1.30 and 1.43 times higher than that of the control group (4.97 ± 0.72). (3) The level of reactive oxygen species (ROS) in the mouse spleen was decreased after 20 and 50 mW laser irradiation, ROS level in the 50 mW group was significantly lower than that of the control group (P 〈 0.05). (4) The percentages of spleen CD3^+ , CD8^+ T cell subpopulations in 20 and/or 50 mW groups and the CD4^+/CD8^+ ratio of peripheral blood in all irradiated groups were decreased significantly ( P 〈 0. 05 ) after irradiation. Conclusions Our results suggest that laser irradiation could not only positively regulate immune function, but also negatively regulate immune function in mice. It provides a new insight into therapeutic mechanism of low energy laser irradiation in clinical application.
