脂质体-c-erbB-2反义寡核苷酸转染对人卵巢癌裸鼠皮下移植瘤形成的抑制作用研究

摘    要:目的 探讨经脂质体—硫代磷酸化修饰的c erbB 2反义寡脱氧核苷酸 (ASODN )作用后的卵巢上皮癌细胞株SKOV3 在裸鼠皮下的成瘤能力。方法 利用脂质体将经硫代磷酸化修饰的寡核苷酸导入SKOV3 细胞内 ,然后将这种被转染的细胞接种于裸鼠皮下 ,观察肿瘤体积的变化 ,并计算抑瘤率。结果 经脂质体 c erbB 2ASODN作用后的卵巢上皮癌细胞在裸鼠皮下成瘤能力降低 ,最大抑瘤率达 4 0 .0 %(P <0 .0 5 )。首次出现目检可见肿瘤的平均时间延长为 9.6天 (P <0 .0 5 )。结论 c erbB 2癌基因的反义调控能降低卵巢上皮癌细胞的成瘤能力 ,抑制肿瘤生长 ,在卵巢上皮癌的基因治疗中有一定价值。

Abstract:Objective To explore the inhibitory effect of liposome|C|erbB|2 antisense phosphorothioate oligodeoxynucleotides (c|erbB|2 ASODN) on the human ovarian epithelial cancer transplanted subcutaneously in nude mice. Methods Human ovarian epithelial cancer cell lines SKOV 3 were treated with liposome |c|erbB|2 ASODN, and then transplanted subcutaneously in nude mice. The changes of tumor volume were observed and the tumor growth inhibitory rate was calculated. Results The tumorigenic ability of the treated cell lines by c|erbB|2 ASODN was reduced, the maximum tumor growth inhibitory rate was 40.0%( P <0.05);The first time that tumor can be detected was raised up to 9.6 days( P <0.05). Conclusion The studies showed that c|erbB|2 ASODN transfection can inhibit the tumorigentsis of ovarian epithelial carcinoma cell lines in nude mice, it may be a valuable gene therapy for the ovarian epithelial carcinoma.;
