复方鳖甲软肝片对5/6肾切除大鼠肾组织 ET-1mRNA表达的影响

摘    要:探讨复方鳖甲软肝片对5/6肾切除(5/6NT)大鼠肾组织内皮素(ET-1)mRNA表达的影响.120只SD大鼠随机均分为正常组,假手术组,5/6NT模型组,苯那普利组,尿毒清组和复方弊甲软肝片大、中、小剂量组.连续用药28周后,处死大鼠,无菌取肾,制作肾组织冰冻制片,用原位杂交方法检测各组ET-1mRNA的表达.结果正常组、假手术组,仅见少量ET-1mRNA阳性反应物;5/6NT模型组,肾小球系膜细胞浆、肾小管上皮细胞浆均见ET-1mRNA强阳性蓝色反应物;苯那普利组、尿毒清组ET-1mRNA阳性反应物减少,与5/6NT组比,P<0.05;复方鳖甲软肝片ET-1mRNA阳性反应物明显减少,并呈剂量依赖关系;与5/6NT组比,P<0.05.结论复方鳖甲软肝片能抑制5/6NT大鼠肾组织ET-1mRNA的表达,这可能是该方防治肾小球硬化的作用机制之一.

Abstract:To study the effect of "Fufang Biejia Ruangan Tablet" on E T-1 mRNA expression in kidney tissue in rats of five-sixths nephretomy (5/6NT) , SD rats were randomized into normal group, sham group, model group, bennapuli group, niaoduqing group, Tablet low-dose group, Tablet medium-dose group and T ablet high-dose group. After 28-week consecutive corresponding treatment, the rats were sacrificed and kidney tissues were taken away to assay the ET-1 mRNA expressions. Results: In normal group and sham group, there were a few ET-1 mRNA positive reactants; in model group, there were more ET-1 mRNA pos itive reactants; in bennapuli group and niaoduqing group, ET-1 mRNA positive re actants reduced, with a difference in comparison with model group (P<0.05); in the three Tablet groups, the ET-1 mRNA positive reactants reduced obviously, with a difference with model group(P<0.05), depending on the dose. Conclusi on: The "Fufang Biejia Ruangan Tablet" may inhibit the ET-1 mRNA expression t o prevent and treat glomerular sclerosis.
