摘    要:目的 探讨端粒酶hTR、hTERT基因表达在人成釉细胞瘤(ameloblastoma,AB)和牙源性角化囊肿(odontognic keratocysty,OKC)中的意义,探讨其在AB、OKC中的发生、发展及其生物学意义。方法 采用mRNA原位杂交方法对口腔颌骨内54例AB、16例OKC及7例口腔正常粘膜,进行了hTR、hTERT mRNA的检测。结果94.4%(51/54)AB有h1、R、hTERTmRNA的表达,81.2%(13/16)、87.5%(14/16)OKC及2/7和1/7正常口腔粘膜分别有hTR、hTERTmRNA表达,3组相比差异有显著性。hTR与hTERT在AB中表达具有相关一致性。端粒酶表达与AB不同组织学类型、病损发生部位、单房与多房无关,在AB外周细胞及星网状多边形细胞中有表达,有角化退变样细胞及颗粒样细胞缺乏表达。结论 hTR、hTERT活化在AB和OKC的发生、发展中起重要作用,有可能成为判断AB预后的可靠指标。

Abstract:Objective To investigate the exprssion of telomrase hTR and hTERT gene in human ameloblastoma (AB) and odontogenic keratocyst (OKC) and to elucidate the genesis,development and biological characteristics of AB and OKC. Methods hTR, hTERT mRNA expression in 54 cases of AB,16 cases of OKC,7 cases of oral normal mucosa was detected by in situ hybridization respectively. Results The positive rates of hTR,and hTERT mRNA in AB were 94.4% (51/54), the positive rates of hTR and hTERT mRNA in OKC, and oral normal mucosa were 81.2% (13/16) ,87. 5% (14/16),2/7 and 1/7 respectively. There was significantly statistical difference. A high correlation between hTERT and hTR mRNA was found. About clinicalpathology of AB, there was no any difference for hTR and hTERT expression, hTR and hTERT mRNA was positive in peripheral and stellate reticula celts. Conclusion hTR and hTERT activity may play an important role in genesis,development of AB. hTR and hTERT may become a new marker for AB prognosis.
