水通道蛋白与脑转移瘤周围水肿的相关性研究

摘    要:目的研究人脑转移瘤及瘤周围脑组织水通道蛋白的表达和调控特征,认识水通道蛋白的异常表达在脑水肿形成和消除中的作用。方法对5例肺腺癌脑转移病例经手术切除的肿瘤组织提取总RNA并进一步行RT-PCR反转录扩增明确脑转移瘤组织中水通道蛋白的表达类型,再对肿瘤组织病理切片行相应水通道蛋白的免疫组化染色,测量切片的灰度值。结果水通道蛋白4(AQP4)在脑转移瘤周围脑组织高表达,而在转移瘤组织内部未见表达。在瘤组织周围,距离瘤组织较远的区域AQP4染色较浅,紧贴瘤组织的区域AQP4染色显著加深。结论AQP4在脑转移瘤周围区域表达上调并以紧贴脑转移瘤的区域为著的现象很可能跟脑转移瘤瘤周脑水肿的形成有关,在脑转移瘤引起的细胞性脑水肿发生机制中发挥重要作用。

Abstract:Objective Study the expression and regulation characteristics of AQPs of cerebuller metastatic tumor and the surrounding of brain tissue tumor.Understand the role of abnormal expression of AQPs in formation and elimination of brain edema.Methods Use extracting total RNA,RT-PCR and immunohistochemical staining to study the expression and regulation characteristics of AQPs with specimens from five lung adencarcinoma brain metastasis cases and measure average gray value.Results AQP4 had a high expression in the peritumoral brain tissue and no expression in the center of brain metastasis tumor organization.Around the tumor tissue,the AQP4 staining was junior in the more distant region from tumor and it added significantly in close to the tumor tissue region.Conclusion The phenomenon that AQP4 had an increased expression in the surrounding region of cerebullar metastatic tumor and moreover,increased significantly in the region next to the cerebullar metastatic tumor tightly is probably related to the formation of peritumoral brain edema and plays an important role in cytotoxic brain edema mechanism.
