摘    要:目的 研究伪狂犬病病毒(PRV)新流行毒株重要毒力基因的分子特征。方法 本研究从江西省某规模化猪场疑似猪伪狂犬病发病仔猪的脑组织中分离到1株病毒,通过PCR鉴定、病毒分离培养、细胞免疫荧光及易感动物试验证实该病毒为PRV野毒株并命名为PRV Jiangxi-FZ株。并对其重要毒力基因TK、gB、gC、gD及gE的分子特征和遗传进化关系进行分析。结果 Jiangxi-FZ株与其他PRV参考毒株的TK、gB、gC、gD及gE基因在核苷酸和氨基酸水平均具有很高的保守性,尤其是与2012年分离的2株PRV变异株的同源性较高。但在高度保守的基础上,仍存在一些差异,且部分差异具有特征性。遗传进化树分析结果显示,PRV Jiangxi-FZ株与2012年国内不同省份分离的PRV变异株亲缘关系较近,而与Becker等欧美洲毒株亲缘关系相对较远。结论 Jiangxi-FZ株具有当前PRV流行毒株的代表性,属近年来流行的PRV变异毒株。

Abstract:To study the molecular characteristics of important virulence genes of Porcine pseudorabies virus (PRV) new epidemic strains, one local strain of PRV was isolated from the brain of a 23-day old pig in Jiangxi Province, and was identified by necropsy, PCR, virus isolation and culture, IFA and animal test. It was named as the PRV Jiangxi-FZ strain. The analysis of molecular characteristic and phylogenetic relationship of its important virulence genes TK, gB, gC, gD and gE showed that the TK, gB, gC, gD and gE gene sequences of Jiangxi-FZ strain shared a strictly conservative with other reference PRV strains at the nucleotide and amino acid levels, especially shared higher homology with 2 PRV variants isolated in 2012. However, there were still some differences between the nucleotide sequences and amino acid sequences of TK, gB, gC, gD and gE genes of Jiangxi-FZ strain and other reference PRV strains on the highly conservative basis, and a part of these differences were characteristic. Phylogenetic analysis showed that the PRV Jiangxi-FZ strain and the PRVs isolated from different provinces in China since 2012 had more closely relationship far away from the European and American PRV strains. Jiangxi-FZ strain had a certain representative of the PRV current epidemic strains, which belong to the PRV variants prevailing in China in recent years.
