摘    要:目的 了解四川省川南地区结核分枝杆菌基因型构成情况,为结核病防控提供分子流行病学依据。方法 采用间隔区寡核苷酸分型(Spacer Oligonucletide typing, Spoligotyping)方法对四川省川南地区267株临床分离菌株进行基因分型,结果采用BioNumerics(Version 5.10)软件进行分析,并将Spoligotyping分型结果与SpolDB4.0数据库中的菌株进行比对分析。结果 267株结核分枝杆菌表现出66种基因型,其中51株表现为独特基因型,其余216株可聚为10个基因簇。与SpolDB4数据库比对分析发现,267株结核分枝杆菌中,144株为北京家族菌株(53.93%),79株为T家族菌株(29.59%),9株为H家族菌株,3株为U家族菌株,2株为MANU家族菌株,其余30株表现的基因型为数据库中不存在的基因型。结论 四川省川南地区结核分枝杆菌存在较高的基因多态性,北京家族菌株和T家族菌株为主要的流行菌株,应加强对这两类菌株的流行趋势的监测及生物学特性研究。

Abstract:We investigated the distribution of genotypes of Mycobacterium (M.) tuberculosis in the South of Sichuan Province. And furtherly, to provide the basis on molecular epidemiology for tuberculosis (TB) prevention and control. In the year of 2010, 267 clinical isolates were genotyped by using spacer oligonucleotide typing (Spoligotyping) in the South of Sichuan. After that, BioNumerics (Version 5.10) software was used to analysis the results and then compared them to the SpolDB4 database. Of the 267 M. tuberculosis isolates, there were 66 genotypes. Of which 51 strains showed unique genotypes, and the remaining 216 strains were grouped into 10 gene clusters. By comparing with the SpolDB4.0 database,we found that 144 strains belonged to the Beijing family (53.93%), 79 strains were the T family (29.59%), nine H-family strains, three U-family strains, two MANU-family strains, the other 30 strains showed strange genotype that didn't exist in the SpolDB4.0 database. In the South of Sichuan, M. tuberculosis has high gene polymorphism, Beijing family and T family strains are the main epidemic strains. It’s necessary to strengthen the monitoring and biological characteristics research of the two genotypes strains.
