槲皮素对糖尿病大鼠肾脏NADPH氧化酶p47phox亚基表达的影响

摘    要:目的探讨槲皮素干预对糖尿病大鼠肾脏NADPH氧化酶p47phox亚基表达的影响及意义。方法采用腹腔注射链脲佐菌素(STZ)方法构建糖尿病模型,32只大鼠分为正常对照组、糖尿病未治疗组、槲皮素小剂量干预组及槲皮素大剂量干预组,12周后称体质量、肾重,计算肾重指数,Western blot检测各组肾脏p47phox、NF-κb、FN和ColIV的表达,应用流式细胞仪检测组织中活性氧簇(ROS)的表达。结果糖尿病未治疗组肾重指数增高(P<0.01);p47phox、NF-κb、FN、ColIV及ROS表达显著上调(P均<0.01)。槲皮素干预能降低肾重指数(P<0.01);减少糖尿病大鼠肾脏p47phox、NF-κb、FN、ColIV及ROS的表达(P均<0.01),以上效应具有剂量依赖性。结论槲皮素干预可以减轻糖尿病大鼠肾病病变,p47phox表达下调导致氧化应激减弱可能是其作用机制之一。

Abstract:Objective To investigate the effects of quercetin on the expression of NADPH oxidase p47phox subunit in the renal lesions of diabetic rats.Methods Diabetes was induced in Sprague Dawley rats with a single intraperitoneal injection of STZ and thirty-two rats were randomized into four groups: normal control,untreated and STZ-induced diabetic rats,and diabetic rats treated with low-dose and high-dose quercetin.After 12 weeks′intervention,body weight,kidney weight index for animals in each group were measured.The expressions of p47phox,NF-κb,FN and ColIV in the kidneys were determined by western blot,and the ROS level in the kidneys was determined by flow cytometry analysis.Results Compared with the normal control group,untreated diabetic group showed significant increases in kidney weight index and the expression levels of p47phox,NF-κb,FN,ColIV and ROS(all P<0.01).Quercetin treatment reduced above-mentioned parameters(all P<0.01) in a dose dependent maner.Conclusions Quercetin treatment can improve renal lesions of diabetic rats.We hypothesize that the expression of p47phox is reduced by quercetin treatment,which leads to a decreased expression of ROS and NF-κb and also results in the reduced expression of FN and ColIV.
