运动对2型糖尿病小鼠骨骼肌基因表达的影响

摘    要:目的利用基因表达谱芯片技术,了解运动对2型糖尿病小鼠骨骼肌基因表达的影响。方法以涵盖14 000个明晰基因的Affymetrix基因表达谱芯片Mouse Genome 430A 2.0分别检测2型糖尿病小鼠力竭运动组和对照组的骨骼肌基因表达谱,利用实时荧光定量PCR方法对芯片差异表达基因进行抽样验证,利用生物信息学方法对检测结果进行分析。结果2型糖尿病小鼠力竭运动组与对照组比较,共筛选出差异表达基因98条,52条基因表达增加,46条基因表达降低。结论利用基因表达谱技术阐明了2型糖尿病小鼠骨骼肌基因表达谱,为进一步研究运动对2型糖尿病的影响提供了依据。

Abstract:Objective To explore the skeletal muscle gene expression profiles of type 2 diabetes mouse by microarray and its alteration after exercise. Methods The skeletal muscle gene expression profiles were tested by Affymetrix Mouse Genome 430A 2.0 gene chip, which approximately included 14,000 well-characterized mouse genes. Real time fluorogenic quantitative RT-PCR method was used to validate the results of the microarray. The results were analyzed by bioinformatical method. Results Among the total 14,000 genes, the expressions of 52 genes were up-regulated and 46 genes were down-regulated in exhausted exercise group when compared with basal metabolism group. Conclusions The results provide basal data for the further research of exercise influence on type 2 diabetes.
