安心颗粒对心力衰竭大鼠细胞凋亡基因Bcl-2及Fas表达的影响

目的探讨安心颗粒防治心力衰竭的作用机制。方法将50只雌性SD大鼠随机分为正常对照组、模型组、卡托普利组、安心颗粒小剂量组、安心颗粒大剂量组,每组10只。采用阿霉素腹腔注射造模,各用药组同时灌胃给药。连续给药6周后,分别用TUNEL法检测大鼠心肌细胞凋亡,用免疫组化法检测大鼠心肌Bcl-2,Fas蛋白的表达。结果安心颗粒能减少心肌细胞凋亡指数,促进Bcl-2蛋白表达,抑制Fas蛋白的过渡表达,其抑制细胞凋亡的作用与卡托普利相当。结论安心颗粒能够上调心肌细胞Bcl-2蛋白、下调Fas蛋白的表达,抑制心肌细胞凋亡,这可能是安心颗粒防治心力衰竭的作用机制之一。

Effect of Anxin Granule on Apoptosis and its Expression of Related Genes Bcl - 2, Fas in Rats with Heart Failure

Objective To study the mechanism of Anxin granule on prevention and treatment of heart failure(HF).Methods Fifty female SD rat were divided randomly into five groups:A(normal control),B(model group),C(captopril control group),D(Anxin granula low-dose group),E(Anxin granula high-dose group)groups.There were 10 rats in each group.Adriamycin was injected to induce HF model.At end of the experiment,the rats were killed,left ventricle tissues were cut to measure cardiocytes apoptosis by TUNEL and the expression of Bcl-2 and Fas proteins in cardiac muscle were tested by immunohistochemistry.Results Anxin granule could decrease the index of cardiocytes apoptosis,enhance Bcl-2 expression and lower Fas expression.The effects of Anxin granule on cardiocytes apoptosis were similar to that of Captopril.Conclusion Anxin granule could up-regulate Bcl-2 expression and down-regulate Fas expression,resisting cardiocytes apoptosis.This might be one of the mechanisms for preventing and treating HF by Anxin granule.
