含酮替芬的罂粟碱液对静脉"桥"血管保护作用的研究

摘    要:目的 探讨并对比分析含富马酸酮替芬的罂粟碱液与常规罂粟碱保存液对冠脉旁路移植术静脉"桥"血管的保护作用,选择能最大限度地保护静脉血管壁内皮细胞(VEC)及血管内膜完整性的保存液,提高其在临床上行冠脉旁路移植术后大隐静脉"桥"血管的远期通畅率.方法 筛选20例行冠状动脉旁路移植术(CABG)的患者,取其CABG术后剩余的大隐静脉(GSV)并分割成2段,常温下分别保存在常规罂粟碱液(对照组)及含富马酸酮替芬的罂粟碱液(实验组)中各1 h.①采用酶联免疫分析法(ELISA法)检测GSV"桥"血管内皮细胞黏附分子-1(ICAM-1)浓度的表达值;②利用高倍光镜观察血管"桥"肌层的肌纤维板、内皮细胞形态结构及单位面积血管内皮细胞的死亡数目;③采用ELISA法检测血管内皮素-1(ET-1)浓度的表达情况.结果 ①实验组ICAM-1浓度的表达值低于常规对照组,光镜下"桥"血管内皮肌层肌纤维板及单位面积VEC的死亡分级中Ⅰ级损伤低于对照组,差异有统计学意义(P<0.05);Ⅱ、Ⅲ、Ⅳ级损伤差异无统计学意义(P>0.05).②实验组内皮细胞ET-1的含量略低于对照组,差异有统计学意义(P<0.05).结论 含富马酸酮替芬的罂粟碱液能够有效地抑制静脉"桥"血管内皮细胞黏附因子的表达,降低单位面积VEC的死亡数目及细胞器的损伤程度.在CABG术中可以选取含富马酸酮替芬的罂粟碱液作为桥血管的保存液,更好地保持术后远期的通畅性.

A study of protection effect on veins "bridge" in contained ketotifenor papaverine liquid

Abstract:Objective To study and analyze two preservation solutions containing Ketotffen Fumarate Papaverine and conventional Ketotifen at the same time in protective effects on the "bridge" graft of coronary artery bypass grafting choose preservation solution that can largely protect the venous wall endothelial cells (VEC) and the vascular intimal integrity to improve its long-term patency of saphenous vein grafts in clinical after coronary artery bypass grafting (CABG). Methods Screening 20 patients with coronary artery bypass grafting (CABG). The remaining saphenous vein after CABG were divided into two sections, respectively stored in conventional poppylye (control group) and contain rich fumaric acid for findan peppylye ketone(group) in each hour at ambient tempera- tures. (1)The enzyme-linked immunoassay assay(ELISA) was performed to measure the concentration of expressio- nendothelial cell adhesion moleculebridge-1 (ICAM-1). (2)Observe the muscularis neoplasm board, endothelial cells, morphology and unit area of endothelial cell death at high magnification. (3)The concentration of endothelin - 1 (ET-1) was measured by using ELISA. Results (1)The expression of ICAM-1 levels in experimental group low- er than that in control group. Death I level in grading of the muscularis neoplasm board and unit area of en- dothelial cell death at high magnification significanflylower than that in control group(P〈0.05). II ,III, IV-level damage was no significant differences (P〉0.05). (2)The expression of ET-I levels in endothelial cells at experi- mental group lower than the control group significantly(P〈0.05). Conclusion Contain rich fumaric acid for findan poppylye testosterone can restrain venous bridge endothelial cell adhesion express, reduce the factor of the VEC per unit area of the number of deaths and organelles. In CABG surgery can select papaverine solution with Ketotifen Fumarate grafts as the preservation fluid to keep the better long-term patency after surgery.
