摘    要: 目的:建立监测重组人粒细胞集落刺激因子(rhG-CSF)复性动态过程的方法?方法:反相高效液相色谱法?采用VydacC4(150mm×4.6mm)柱,流动相为三氟醋酸-乙腈混合物梯度洗脱?结果:发现完全复性有生物学活性的rhG-CSF在25.4min左右出峰,而完全变性无生物学活性的rhG-CSF在26.4min左右出峰,两峰出峰时间相差1min左右?随着复性时间的增加,两者的峰面积之比在变化?Tris复性体系与PB复性体系相比,复性收率差不多,都可在12h内达到的70%以上复性正确率?结论:此方法适用于基因工程产品纯化过程中复性产率的控制?

Abstract:OBJECTIVE:To develop an assay in order to monitor the kinetic processes of refolding of recombinant human granulocyte colony stimulating factor.METHOD:A reversed-phase high-perfermance liquid chromatography was developed.Vydac C4(150 mm×4.6 mm) column was used and the mobile phase consisted of a mixture of trifluoroacetic acid-acetonitrile.RESULTS:The correct folding peaks and denatured peaks were separated well.The peak appeared on 25.4 min had biology activity.CONCLUSION:This method may be used for the measurements of correct folding rate of rhG-CSF.
