HGF、IL-6在实验性早期肝癌中的表达及其诊断意义

目的：探讨肝细胞生长因子（HGF）、白细胞介素－6（IL－6）在实验性早期肝癌中的诊断意义。方法：用ELISA、免疫组化、免疫电镜方法分别检测HGF及IL－6在实验组及正常对照组的血清水平和在肝细胞中的表达情况，用免疫细胞化学方法检测HGF在人肝细胞系中的表达。结果：HGF、IL－6在肝癌旁组织肝细胞质中表达阳性。肝癌组织中表达均下降，人肝癌细胞系中HGF强阳性表达，实验组HGF、IL－6血清含量显著高于正常对照组（P＜0．01）。结论：化学致癌剂诱发实验性早期肝癌过程中、IL－6可能是激活HGF的细胞因子，HGF以自分泌机制发挥促肝细胞增殖的效用。HGF、IL－6有望成为早期肝癌的诊断指标。

Expression of HGF, IL-6 in early experimental liver carcinomas and their significance in early diagnosis

Purpose To investigate the diagnostic significance of hepatocyte growth factor(HGF), interleukin 6(IL 6) in early experimental hepatocellular carcinomas. Methods The serum levels of HGF, IL-6 were measured by ELISA. The expression of HGF, IL 6 in the liver tissues of rat early experimental hepatocellular carcinomas and human hepatoma cell lines were studied by immunohistochemistry, immunocytochemistry and electron microscopic immunocytochemistry. Results HGF, IL 6 were expressed in the cytoplasm of hepatocytes in paracancerous liver cirrhotic tissues and their expression were decreased in early experimental hepatocellular carcinoma tissues. HGF still was expressed in human hepatoma cell lines. The both of HGF, IL 6 serum level of rat early experimental hepatocellular carcinomas were higher than those of the control group significantly( P <0 01). Conclusions During rats are induced into early hepatocellular carcinomas by chemical carcinogen,the gene of HGF may be actived by a cytokine, IL 6. An autocrine mechanism was suggested in HGF for the proliferation of hepatocytes. HGF and IL 6 are possible biological markers in early diagnosis of hepatocellular carcinoma.
