慢性低氧性肺动脉高压大鼠体内血管活性物质表达的变化及意义

摘    要:目的: 观察慢性低氧性肺动脉高压(HPH)大鼠血浆中心房钠尿肽(ANP)和降钙素基因相关肽(CGRP)的表达变化.方法: 用低压、低氧的方法复制大鼠慢性HPH模型,放射免疫法检测血浆中血管活性物质ANP和CGRP表达的变化.结果: 慢性HPH大鼠血浆中血管活性物质ANP和CGRP的表达均发生了变化,呈现先降低后增高的表现, 即低氧3 wk>正常>低氧2 wk.结论: 在慢性HPH形成过程中,随着肺小动脉壁的增厚,血浆中血管活性物质ANP和CGRP的表达先降低后增高.其变化可能与慢性HPH过程中肺动脉压升高和肺小动脉壁的结构重建有一定关系.

Abstract:AIM: To observe the expression of such vasoactive peptides as atrial natriuretic peptide (ANP) and calcitonin gene-related peptide (CGRP) in the plasma of rats with chronic hypoxic pulmonary hypertension (HPH). METHODS: Twelve chronic HPH rat models were established in low-pressure and hypoxic conditions and divided evenly into 2 groups: hypoxic 2-week group and hypoxic 3-week group. Another 6 rats as control group were fed under normal pressure and normoxia conditions. The expressions of ANP and CGRP in the plasma of HPH rats were detected by radioimmunoassay (RIA) method. RESULTS: RIA results showed that compared with the concentrations of ANP and CGRP in plasma of control group, the concentrations of ANP and CGRP in plasma of hypoxic 2-week group decreased while those of hypoxic 3-week group increased. The tendency was hypoxic 3-week group>control group>hypoxic 2-week group. Compared with those in hypoxic 2-week group, the expressions of ANP and CGRP were higher in hypoxic 3-week group (P< 0.05) . CONCLUSION: In the process of chronic HPH, the expressions of ANP and CGRP in plasma decrease first and then increase . ANP and CGRP may play a regulative role in pulmonary hypertension and pulmonary arterial reconstruction in chronic HPH rats. There is a correlation between the low-pressure and hypoxia induced HPH and the increased ANP and CGRP in rats.
