摘    要:目的 探讨温和性高温对鼻咽癌细胞系CNE-2Z放射增敏作用的机制.方法 实验分4个组,对照组、单纯加热组、单纯照射组和联合处理组.采用流式细胞仪分析G2/M期分布比例及凋亡百分比,Western-Blotting检测CyclinB1的表达,改良的荧光染色法观察核碎片形态.结果 联合处理组G2/M期分布比例较高,CvclinB1表达较高,并产生更多的核碎片,放射诱导的凋亡百分比并不增加.结论 G2/M 期分布比例增高,CyclinB1表达增加是温和性高温对鼻咽癌细胞系CNE-2Z放射增敏作用的机制.

Abstract:Objective To study the mechanism of radiosensitization of hyperthermia on nasopharyngeal carcinoma cell line CNE-2Z. Methods 4 groups were involved in the experiment: the control group, the simple heat group, the simple radiation group and heat plus radiation group. The change of the distribution of G2/M phase was analyzed by the flow cytometry (FCM), and proportion of apoptosis in addition. Western-Blotting was used to detect the expressions of CyclinBl. Microscope was used to observe the morphology of cells stained with Hoechst 33342. Results More distribution of G2/M phase, along with higher expression of cyclinB1, was found in the heat plus radiation group,leading to more nuclear fragments. Hyperthermia did not add to more apoptosis. Conclusions More distribution of G2/M phase, along with higher expression of cyclinB1 might be the mechanism of radiosensitization of hyperthermia on nasopharyngeal carcinoma cell line CNE-2Z.
