摘    要:目的建立高效液相色谱法测定人血浆中伏立康唑的含量。方法血浆样品经乙酸乙酯-环己烷萃取,以双嘧达莫为内标,采用AgilentZORBAXSB-C18色谱柱(4.6mm×150mm,5μm)为分析柱,以三乙胺-冰醋酸-水-乙腈(1:1:65:35)为流动相,检测波长为254nm,柱温为30℃,流速为1.0ml/min。结果伏立康唑及内标双嘧达莫的色谱峰分离良好,伏立康唑在0.1~10μg/ml范围内线性关系良好,r=0.9981(n=6),平均方法回收率为93.55%,RSD小于2%,日内日间RSD均小于15%,定量限为0.1μg/ml。结论本方法简便快速,准确灵敏,适用于人血浆中伏立康唑的血药浓度监测。

Abstract:Objective To establish an high performance liquid chromatography method for the determination of voriconazole in human plasma.Methods The analytes in plasma were extracted by acetoacetate and hexamethylene.Using dipyridamole as the internal standard,voriconazole was separated on Agilent ZORBAX SB-C18(4.6 mm×150 mm,5 μm).The mobile phase consisted of a mixture of triethylamine,acetic acid,water and acetonitrile (1:1:65:35) at a flow rate of 1.0 ml/min.The detection wavelength was 254 nm,and the column tempreture was 30℃.Results Voriconazole was well separated.The calibration curve was linear in the range of 0.1-10 μg/ml,with a correlation coefficient of 0.9981(n=6).The average recovery was 93.55% and the relative standard deviation(RSD) was below 2%.The intra-day and inter-day RSD was less than 15%.The limit of detection(LOD) was 0.1 μg/ml for voriconazole.Conclusion The method is accurate and simple when used for the determination of voriconazole in human plasma.
