维生素D受体异常表达对大鼠肾小管上皮细胞增殖凋亡的影响

摘    要:目的 研究维生素D受体(VDR)异常表达对大鼠肾小管上皮细胞增殖凋亡的影响。方法 制取大鼠肾小管上皮细胞,并构建VDR干扰和过表达载体。将VDR干扰和过表达载体分别转染到大鼠肾小管上皮细胞中,通过MTT法检测VDR干扰和过表达后大鼠肾小管上皮细胞的增殖;流式细胞术检测细胞周期;Annexin - PI法检测细胞凋亡情况。结果 与对照组相比,过表达VDR后,大鼠肾小管上皮细胞的VDR蛋白表达显著上调,增殖速度下降,发生G1期阻滞,凋亡比例增多,差异具有统计学意义。而干扰VDR表达后,结果与上述相反,即大鼠肾小管上皮细胞的VDR蛋白表达显著下调,增殖速度加快,细胞凋亡比例降低,差异具有统计学意义。结论 VDR能促进大鼠肾小管上皮细胞发生细胞周期G1期阻滞,抑制细胞增殖,促进细胞凋亡。

Abstract:Objective To investigate the effects of abnormal Vitamin D receptor(VDR) expression on the proliferation, cell cycle and apoptosis of rat renal tubular epithelial cells. Methods Rat renal tubular epithelial cells were obtained, then the interference and overexpression vectors of VDR were constructed.VDR interference and overexpression vectors were transfected in to rat renal tubular epithelial cells respectively. Tenal tubular epithelial cells proliferation was detected by MTT assay. Cell cycle was detected by flow cytometry and apoptosis index were detected by Annexin-PI assay after the interference and overexpression of VDR. Results Compared with control group, theexpression of VDR protein in rat renal tubular epithelial cells significantly up-regulated after overexpression of VDR, proliferation rate decreased, G1 phase arrest occurred, and the apoptotic rate increased. The difference was statistically significance. However, after interfering with the expression of VDR, the results were contrary to those mentioned above, the expression of VDR in rat renal tubular epithelial cells was significantly down-regulated, the proliferation rate was accelerated, and the percentage of apoptotic cells was significantly decreased. The difference was statistically significance. Conclusion VDR can promote G1 phase block, inhibit cell proliferation and promote cell apoptosis of renal tubular epithelial cells.
