南京市一起斯坦利沙门氏菌食物中毒的病原学检测及溯源分析

摘    要:目的 对食物中毒事件中所采集样本进行食源性致病菌的分离鉴定,快速查明食物中毒原因并进行同源性分析。方法 应用实时荧光PCR技术对一起食物中毒10份可疑食品、2份餐具涂抹样和5份患者粪便进行检测,同时进行病原菌培养分离鉴定,分离菌株采用CLSI 推荐的改良K-B纸片法进行抗生素敏感试验,应用PFGE分型技术进行同源性分析。结果 1份可疑食物和3份患者粪便经实时荧光PCR检测为沙门菌阳性,同时分离到4株斯坦利沙门氏菌,4株沙门氏菌的药敏试验结果一致,PFGE 带型完全一致。结论 本次食物中毒是由斯坦利沙门氏菌污染引起。实时荧光PCR技术能够快速准确地对食物中毒做出病原学诊断,PFGE基因分型技术可对病原菌进行溯源分析,对研究病原菌流行病学规律具有重要意义。

Abstract:Objective To isolate and identify food-borne pathogens in samples collected from food poisoning events,and to quickly identify the causes of food poisoning and to conduct homology analysis.〖WTHZ〗Methods Real-time fluorescent PCR was used to detect 10 food suspected food poisons,2 tableware spreads and 5 patient stools.The pathogen culture was isolated and identified.The isolates were tested by CLSI-recommended modified KB paper for antibiotic sensitivity test.The PFGE typing technique was used for homology analysis.〖WTHZ〗Results One suspected food and three patients′ stools were positive for Salmonella by real-time fluorescent PCR.Four strains of Stanley were isolated.The results of four strains of Salmonella were consistent and the PFGE bands were identical.〖WTHZ〗Conclusion The pathogen for this food poisoning was Salmonella Stanley.Real-time fluorescent PCR technology can quickly and accurately determine the pathogens of food poisoning.The PFGE has provided a useful tool for bacterial strains homology and traceability analysis of pathogens,and is of great significance for the epidemiology of pathogens.
