孕哺期氧化乳蛋白饮食对子代小鼠学习记忆能力的影响

摘    要:目的 探索孕哺期氧化乳蛋白饮食对子代小鼠脑部氧化还原状态及学习记忆能力的影响。方法 通过不同处理方法加热、次氯酸(HClO)、过氧化氢-铜(H2O2-Cu)]对全脂乳进行氧化修饰。雌雄鼠合笼成功即开始饲喂氧化乳蛋白日粮3w,产后母乳喂养,仔鼠3w断乳后,随机分为三批,第一批立即测定仔鼠脑部的氧化损伤指标,第二、三批分别饲以原来氧化乳蛋白日粮和普通饲料至6w,进行Morris水迷宫测试空间学习记忆能力;测定6w仔鼠脑部的抗氧化及氧化损伤指标。结果 孕哺期饲喂氧化乳蛋白日粮,子代小鼠脑部氧化还原状态失衡,与空白对照组相比,氧化修饰组子代小鼠脑部抗氧化酶活力(CAT、SOD)和总抗氧化能力(T-AOC)均呈现不同程度的下降(P<0.05),水迷宫实验中,与空白对照组相比,氧化乳蛋白组潜伏期明显上升,穿越平台次数低于空白对照组(P<0.05)。结论 孕哺期饲喂氧化乳蛋白日粮,引起子代小鼠脑部氧化应激,降低其抗氧化防御能力,同时影响其空间学习记忆能力。

Abstract:Objective To explore the brain redox state effects and spatial learning and memory of generation mice treated with oxidation dairy protein during pregnancy and lactation period.〖WTHZ〗Methods Whole milk were modified respectively by heat,HClO and H2O2-Cu.Oxidized dairy protein was fed after the female and male rats were caged successfully.The generation of mice were breast-fed after birth.After weaning at 3 weeks old,the generation mice were randomly divided into three groups,the first group were immediately measured for oxidative damage on the brain,the second group were fed with the oxidation of dairy protein to 6 weeks and the third group were fed with ordinary feed to 6 weeks.Morris water maze task was applied to evaluate spatial learning and memory in the generation mice.Antioxidant and oxidative damage index of the mice were detected.〖WTHZ〗Results Comparing to the control group,in brain of generation mice fed with oxidation of dairy protein during pregnancy and lactation,activities of anti-oxidant enzymes (SOD,CAT) and T-AOC significantly decreased (P<0.05),moreover,the incubation period increased significantly and the number of times of crossing the platform was lower than the control group (P<0.05).〖WTHZ〗Conclusion Oxidation dairy protein during pregnancy and lactation would lead to oxidative damage on the brain of generation mice,and it also affects their spatial learning and memory ability.
