摘    要:目的 建立反相高效液相色谱法测定人血清中万古霉素(糖肽类抗菌药)的浓度.方法 色谱柱为Shimadzu Shim-pack VP-ODS柱,流动相为甲醇-乙腈-0.05 mol·L-1磷酸二氢钾缓冲液(pH 3.2)=6.5:8.5:85,流量0.8 mL·min-1,紫外检测波长236 nm;盐酸去甲万古霉素为内标.结果 血清万古霉素在1-100 mg·L-1内线性良好,回归方程Y=24.09x-0.08(n=7,y=0.9993),日内和日间精密度RSD<10%.结论 该测定方法可靠,可用于临床血清万古霉素的浓度监测.

Abstract:Objective To establish a method of determining serum vancomycin in human by RP - HPLC. Methods Separation was car-ried out on a Shimadzu Shim -pack VP -ODS column, the mobile phase consisted of methanol, acetonitrile and buffer ( 6.5:8.5:85 ),flow rate was 0.8 mL·min-1 , UV detector wavelength was 236 nm. Norvancomycin was used as internal standard. Results A good lineari-ty was demonstrated between 1-100 mg·L-1 by linear equation y =24.09x - 0.08 (n=7,γ= 0.9993 ). The intra - day and inter - day deviation was showed by RSD < 10%. Consulsion The method is val-idated for determination of serum vancomyc in clinical practice.
