叉头框转录因子M1在甲状腺乳头状癌组织中的表达及意义

摘    要: 目的探讨叉头框转录因子M1(FoxM1 )在人类甲状腺乳头状癌组织中的表达及其意义。方法利用实时荧光定量PCR、Western blot检测20例甲状腺乳头状癌组织及相应的20例癌旁正常组织和20例甲状腺腺瘤组织中FoxM1 mRNA和蛋白的表达水平并进行统计学分析。结果FoxM1 mRNA及蛋白表达在甲状腺乳头状癌组织中显著高于癌旁正常甲状腺组织及甲状腺腺瘤组织(均P <0.01),癌旁正常甲状腺组织和甲状腺腺瘤组织比较差异无统计学意义(P >0.05),甲状腺乳头状癌有淋巴结转移组高于无淋巴结转移组(P <0.01),包膜侵犯组高于包膜无侵犯组(P <0.01)。FoxM1 mRNA及蛋白表达与患者的性别、年龄、肿瘤大小及TNM分期均无关(P >0.05)。结论FoxM1 mRNA及蛋白的异常表达可能与甲状腺乳头状癌的发生发展、淋巴结转移、包膜侵犯有关,有望成为甲状腺乳头状癌诊治中的一个新的标志物。

Abstract:Objective To investigate the expression and clinical significance of FoxM1in human papillary thyroid carcinoma. Methods Quantitative real-time PCR and Western blot was performed to detect the mRNA and protein expressions of FoxM1in papillary thyroid carcinoma tissues (20 cases),adjacent normal tissues (20 cases),and thyroid adenoma (20 cases); statistical analyze was performed. Results The expressions of FoxM1mRNA and protein in papillary thyroid carcinoma were significantly higher than that in adjacent normal tissues (P<0.01) and in thyroid adenoma tissues (P<0.01). There was no significant difference in expression of FoxM1between adjacent normal tissues and thyroid adenoma (P>0.05). The expressions of FoxM1mRNA and protein were higher in the group with lymph node metastasis than those without lymph node metastasis (P<0.01),and the expressions of FoxM1mRNA and protein were higher in the group with capsule invasion than in the group without capsule invasion (P<0.01). The expressions of FoxM1mRNA and protein in papillary thyroid carcinoma were not correlated to patients? gender,age,tumor size or TNM stage (P>0.05). Conclusions The abnormal expression of FoxM1may be associated with the oncogenesis and progress of papillary thyroid carcinoma and lymph node metastasis,capsule invasion,which is expected to become a new marker for diagnosis and treatment of papillary thyroid carcinoma.
