白花前胡丙素对实验性心肌缺血损伤的影响

摘    要:目的:观察白花前胡丙素对小鼠耐缺氧能力的影响以及对连二亚硫酸钠(Na_2S_2O_4)和过氧化氢(H_2O_2)所致幼鼠心肌细胞损伤的保护作用。方法:采用异丙肾上腺素制备低氧模型,记录小鼠缺氧条件下的存活时间;应用原代培养的乳鼠心肌细胞,用Na_2S_2O_4和H_2O_2制备心肌缺血性损伤模型,分别测定各组心肌细胞的存活率(MTT法)、乳酸脱氢酶(LDH)和超氧化物歧化酶(SOD)含量。结果:白花前胡丙素(10,20 mg·kg~(-1))明显延长异雨肾上腺素所致小鼠缺氧条件下存活时间(P<0.01);白花前胡丙素(20,40,80 mg·L~(-1))降低细胞死亡率,显著抑制H_2O_2和Na_2S_2O_4损伤引起的心肌细胞LDH释放,并提高细胞中SOD浓度(P<0.05,P<0.01);对正常生长条件下的心肌细胞无明显影响(P>0.05)。结论:白花前胡丙素对缺氧小鼠及缺氧心肌细胞有明显的保护作用,这可能与其抑制脂质过氧化反应有关。

Abstract:AIM:To observe the protective effects of praeruptorin C (Pra-C) against hypoxic mice and the cardiomyocytes injuried by sodium hydrosulfite and hydrogen peroxide.METHODS:The hypoxic animal models were observed by means of injecting isoprenaline 20mg·kg~(-1) and the survival time of the mice model under the hypoxic condition was recorded.The cultured neonatal rat cardiomyocytes were injured by sodium hydrosulfite and hydrogen peroxide,the activity,the content of dehydrogenase(LDH),and surperoxide dismutase (SOD) of cardiomyocytes in the medium were measured separately.RESULTS:Pra-C (10 and 20mg·kg~(-1))could significantly prolong the survival time in myocardial hypoxie mice induced by isoprenaline. Pra-C(20,40,80 mg·L~(-1))could decrease the mortality of the cardiomyocytes,distinctively inhibite release of LDH,and increase SOD of cardiomyocytes injured by H_2O_2 and Na_2S_2O_4 (P<0.05,P<0.01).Pra-C has no effect on cardiomyocytes under normal condition (P>0.05).CONCLUSION:Pra-C shows remarkable protective effects on myocardial hypoxic mice and cardiomyocytes,which maybe related to its inhibition of lipid peroxidation.
