CD4^+CD25^+CD127low调节性T细胞及相关细胞因子在儿童再障发病中的作用

摘    要:目的探讨CD4^+CD25^+CD127low调节性T细胞及转化生长因子-β1(TGF-β1)在再障患儿外周血中的表达及临床意义。方法采用流式细胞术分别检测21例慢性再障(CAA)、9例急性再障(SAA)和15例正常健康儿童外周血自细胞,并用酶联免疫吸附法检测其外周血TGF-β1表达水平。结果外周血中CD;T细胞百分比、CD4^+CD25^+,CD4^+CD25^+CD127low调节性T细胞占CD4^+T细胞百分比及血清TGF-β1表达水平再障患儿均低于正常健康儿童(P均〈0.05),S从低于CAA(P均〈0.05);再障患儿CD4^+CD25^+CD127low调节性T细胞占CD4^+T细胞百分比与TGF-β1的表达水平无明显相关性。结论再障患儿周血调节性T细胞比例减少、血清TGF-β1表达水平减低可能参与了再障的发病。

Abstract:Objective To explore the levels of CD4^+CD25^+CD127low regulatory T cells (Tregs) and TGF-β1 in peripheral blood of aplastic anemia (AA) in children, and to investigate the role of Tregs in the pathogenesis of aplastic anemia. Methods A total of 21 patients with chronic aplastic anemia (CAA), 9 patients with acute aplastic anemia (SAA) and CD4^+CD25^+CD127low T ceils in CD4+ T cells was evalu-15 normal healthy children were included in this study. The proportion of CD4+CD25^+CD127^low ated by flow cytometric analysis. TGF-β1 was measured by ELISA. Results The percentage of CD4+ T cells, CD4+CD25^+ T cells. Tregs in peripheral blood and serum TGF-β1 expression in patients with AA were lower than those in the normal control (all P 〈 0.05). The percentage mentioned above in SAA was lower than that in CAA (all P 〈 0.05 ). Tregs percentage and the level of TGF-β1 in children with AA were no significant correlation. Conclusions The low percentage of regulatory T cells and the low level of TGF-β1 in children may be involved in pathophysiology of AA.
