骨髓间充质干细胞对自然衰老小鼠T细胞的调节作用研究

摘    要:目的:研究探讨小鼠骨髓间充质干细胞对自然衰老小鼠T细胞的节作用影响情况。方法:通过流式细胞术研究对比衰老小鼠和年轻小鼠T细胞培养48h后CD8+CD28+的共表达率差异,根据以上研究结果分别对比隔离共培养组、混合共培养组和对照组之间T细胞上CD8+CD28+的共表达率差异。结果:实验一中衰老组培养48h后CD8+CD28+共表达率为(4.08±0.3493)%,年轻组培养48h后CD8+CD28+共表达率为(5.04±0.5741)%。两者相比有统计学差异(P〈0.05)。实验二隔离共培养组中骨髓间充质干细胞与淋巴细胞以1:1在Transwell系统中共培养,48h后其对应的CD8+CD28+共表达率为(10.26±1.0784)%,混合共培养组中骨髓间充质干细胞与淋巴细胞以1:1比例在六孔板中共培养,48h后CD8+CD28+共表达率为(6.42±1.5680)%。对照组中淋巴细胞以106数量进行培养,48h后CD8+CD28+共表达率为(4.08±0.3493)%。两两对比有统计学差异(P〈0.05)。结论:衰老小鼠和年轻小鼠的T细胞在CD8+CD28+的共表达比例上存在差异。骨髓间充质干细胞能够通过分泌的某些细胞因子使这种衰老的T细胞变得趋向于"年轻态"。

Abstract:Objective This is a study on effect of mouse BM-MSCs on aging mouse T cell regulation.Methods There were two groups in experiment one,aging group and young group,the difference of CD8+ CD28+ expression between the two groups were detected with flow cytometry,according to the result of experiment one,the difference of CD8 CD28 expression of separated co-culture group,mixed co-culture group and control group,which were called experiment two,was detected with flow cytometry.Results In the aging group of experiment one,the CD8+ CD28+ expression was(4.08±0.3493)%.the CD8+ CD28+ expression of young group was(5.04±0.5741)%.There was a statistical difference.In the separated co-culture group of experiment two,we co-cultured BM-MSCs and lymphocyte in accordance with the proportion of 1:1,and the CD8+ CD28+ expression was(10.26±1.0784)% after 48 hours.In the mixed co-culture,we mixed BM-MSCs with lymphocyte in 6 wells plate in accordance with the proportion of 1:1,and the CD8+ CD28+ expression was(6.42±1.5680)% after 48 hours.In control group,the lymphocyte was clutured alone,and the CD8+ CD28+ expression was(4.08±0.3493) % after 48 hours.There was a statistical difference.Conclusion There were difference of CD8+ CD28+ expression between aging mouse and young mouse.BM-MSCs can take aging T cells to young state.
