| 组织检材的PCR性别鉴定研究 |

| 摘 要： | 作者用引物Ｙ３、Ｙ４通过ＰＣＲ技术对微量陈旧的组织检材进行了扩增，扩增的靶序列位于Ｙ染色体ＤＮＡ特异３．４Ｋｂ重复序列中，产物为４４６ｐｂ。检材是新鲜组织、甲醛固定组织、石蜡包埋组织和ＨＥ染色切片组织。检材的保存时间从凡小时到五年。结果表明：新鲜组织，１０％中性缓冲甲醛固定一年以下的组织和石蜡包埋３年以下的组织均可做为ＰＣＲ性别鉴定的良好检材。

| Abstract: | DNA extracted from the trace and old
specimen was amplificated with prinier Y3、Y4by PCR. The target locus amplificated located
specific sequences一3. 4 Kb tandem repeats in Y chomo-some,the amplificated product was
446 bp. Tissue specimens were fresh ,formalin一fixed and paraffin一embedded,and they were
HE stained section tissue. The stored time were from several hours to fiveyears. The results
suggest that the fresh tissue,10%buffered neutral formalin一fixed tissue for oneyear or less
and paraffin一embedded tissue for three years or less are suitable specimens for sex
identi-fiation by PCR |
