白英提取液对Hela细胞凋亡及P53和Bcl-2蛋白表达的影响

摘    要:目的探讨白英提取液对Hela细胞凋亡及野生型P53(wtP53)和Bcl-2蛋白表达的影响。方法采用MTT法检测细胞增殖抑制率,荧光显微镜检测癌细胞形态变化,TUNEL法检测凋亡率,二步法免疫组化检测P53和Bcl-2蛋白表达变化。结果白英提取液能够抑制Hela细胞增殖,且呈剂量依赖性;观察到凋亡细胞典型的形态特征;细胞凋亡率随白英提取液浓度增加而升高;P53蛋白表达显著上升(P<0.01),而Bcl-2蛋白表达明显下降(P<0.01)。结论白英提取液能促进Hela细胞凋亡,其分子机制可能与上调野生型P53蛋白表达和下调Bcl-2蛋白表达有关。

Abstract:Objective To outline the effects of Solanum lyratum Thunb(ST) extract on induction of apoptosis and the expression of wild type P53 and Bcl-2 protein in Hela cells.Methods A MTT assay was used to detect the cells proliferation inhibitory rate.Morphological changes of apoptosis were observed with fluorescence microscope.Induction of cell apoptotic rate was determined by TUNEL method.The expression of P53 and Bcl-2 proteins were detected by two-step immunohistochemical staining. Results ST extract could inhibit the proliferation of Hela cells and the inhibitory effect was dose-dependent.Typical morphology changes was found in apoptotic cells.The rate of apoptosis increased significantly with the increased concentration of the drug.The expression of P53 protein elevated significantly(P<0.01),while Bcl-2 protein decreased markedly(P<0.01).Conclusion ST can induce the apoptosis of Hela cells,and its molecular mechanism may be associated with the up-regulating expression of wild type P53 protein and down-regulating expression of Bcl-2 protein.
