分离纯化肾小管基底膜的改进方法

摘    要:本实验采用分样筛、磁性分离和密度梯度离心综合法分离猪肾皮质的肾小管基底膜。相差显微镜观察肾小管的纯度可达97%以上。磁性分离后用超声震碎细胞核,再经密度梯度离心,即可获得高纯度的肾小管基底膜。

Abstract:A combined method of graded sieving, magnetic separating and density gradient centrifugating procedure wae used to isolate tubular basement membrane from renal cortex of pigs. The degree of purity of the tubules thus prepared was above 97% by using phase microscopy. After magnetic separation of the tubules,the nuclei were disrupted in a ultrasonicator,and finally the highly puri-fied tubular basement was obtained by discontinuous sucrose density gradient centrifugation.
