养心通脉片对家兔动脉硬化血管中细胞周期蛋白E及其依赖性激酶2的影响

目的：观察细胞周期蛋白E（Cyclin E）及其依赖性激酶（CDK）2在家兔动脉粥样硬化（AS）血管组织中的表达及养心通脉片的干预作用。方法：参照“高脂饲养喂饲法”造模，设养心通脉片组和辛伐他汀片组、模型组（高脂饮食喂养）和健康对照组，分别用养心通脉片、辛伐他汀片稀释液及蒸馏水灌胃。30d后，采用免疫组化法检测四组血管组织中Cyclin E和CDK2的表达强弱。结果：养心通脉片组Cyclin E和CDK2的表达明显低于模型组（P〈0．01或〈0．05），而与健康对照组、辛伐他汀片组比较差异无显著性；但辛伐他汀片组Cyclin E和CDK2的表达显著高于健康对照组（P〈0．05）。结论：动脉粥样硬化与Cyclin E和CDK2表达增加有关，养心通脉片能调节Cyclin E和CDK2的表达，从而可能抑制细胞的异常增殖，有效地逆转或消退动脉粥样硬化斑块。

Effect of Yangxin tongmai tablets on the expression of cyclin E and cyclin dependent kinase 2 in rabbit atherosclerosis model

Objective:To investigate the effect of yangxin tongmai tablets(YTT) on the expression of cyclin E and cyclin dependent kinase 2(CDK2) in the vascular intima of rabbit atherosclerosis models.Methods:New zealand rabbits were divides into four groups: Yangxin tongmai group,simvastatin group,model group and healthy control group,then fed with high-fatty diet except healthy control group.After 30 days,all rabbits were killed and the aorta was dissected,the expression of cyclin E and CDK2 were detected by immunohistochemistry assay.Results:The expression of cyclin E and CDK2 in yangxin tongmai group was lower than those in model group(P<0.01 or P<0.05),but there was no significant difference compared with healthy control group and simvastatin group(P>0.05).Furthermore,compared with the healthy control group,the expression of cyclin E and CDK2 was higher in simvastatin group(P<0.05).Conclusion:The result indicated that the expression of cyclin E and CDK2 is enhanced in atherosclerosis,yangxin tongmai tablets can downregulate its expression,possibly result in the inhibition of vascular smooth muscle cell proliferation and attenuation of the atherosclerotic plaque.
